Study On The Antidiarrhea Effect Of Elaeagnus Angustifolia L. And It's Preparation

Objective: To investigate the antidiarrhea effective part and chemical components of Elaeagnus angustifolia L, through the pharmacodynemics study, verify the extracting technology of effective components and its activities based on the results of pharmacodynemics and prepare the granules. Methods: (1) diarrhea model in mice were induced by senna and castor oil, respectively, the ink gastrointestinal propellant test, propellant augmentation of small intestine test induced by neostigmine were applied. Diarrhea exponent, loose stools rate, small intestine propellant rate were assayed as index, the effects those of different extracts and powder of fruits on the models and the indexes were observed. (2)The alcohol were used as solvent to extract the total tannin, were assayed by the method of ch.p . The activity of total tannin was verified by the models and testes above, respectively. (3)The technology on extracting total tannin were optimized by using orthogonal design based on the contents of total tannin, different concentration ofβ-CD were added to tannin extract to improve sticking phenomenon. The granules formulation and its preparing technology were screened using the rates of forming, moisture and solubility as indexes. (4)The effectiveness of granules prepared were tested using the models and testes above, in addition, the ant-inflammation and analgesic effect of granules were also investigated using the models induced by dimethyl benzene, writhing response induced by acetic acid, respectively. Results: (1)The water extracts ,alcohol extracts and powder can reduce the diarrhea exponent, loose stools rate of mouse diarrhea induced by senna and castor oil, inhibited the mouse normal and augmentation gastrointestinal movement(P<0.05). (2)The contents of total tannin in alcohol extract was 8.23%, the activity of total tannin was better than alcohol extract.. (3)The amount of powder obtained was the most, when 5% ofβ-CD added to. The total tannin extract. It is showed the best rates of forming and moisture when using the lactose and mannitol as excipients. The optimized preparing technology on granules were:70% of ethanol as moisture, one part of extract mixed with two part of excipient (lactose:mannitol=4:1,w/w), to make the soft material, screening and granuling, drying at 80℃, the granule dried were sieved through the 20 meshes and ordered, packed. (4)The effectiveness of granules showed the same results as above, and also inhibited swelling of ear induced by dimethyl benzene(P<0.05), writhing response induced by acetic acid.(P<0.05). Conclusion: Elaeagnus angustifolia L from Xinjiang showed the anti-diarrhea effectiveness in mice and the effective part was total tannin, the corresponding parameters of granules accord with the standard.
