Elaeagnus angustifolia is a deciduous trees of Elaeagnaceae. In our country, mainly in the northwestern provinces, It distributed in 34 ° north. Elaeagnus angustifolia is a woody plant food. Elaeagnus angustifolia’s flowers, fruits, branches, leaves and bark and other parts can be used as medicine. It can be used to treat chronic bronchitis, closed fracture, dyspepsia, neurasthenia and other diseases, Elaeagnus angustifolia is a traditional remedy. This paper aims to test Elaeagnus angustifolia polysaccharide(EAP)for its isolation, purification and properties of the active has done preliminary exploration, provide a basis for the future use and development for EAP.Test to Elaeagnus angustifolia for the study, EAP extraction, purification, antioxidant activity were studied. Are summarized as follows:1.Use single factor test as basis, then use response surface methodology to optimization the extraction process. After use single factor test to ensure response surface methodology conditions range, run 17 experiment to determine the best extraction range. Then use Design Expert 8.0.6 Trail to analysis. Know the best condition is: 61 ℃, extraction time 118.4 min, Raw material and solution’s ratio is 1:26. Under optimum conditions determined experimentally Elaeagnus angustifolia polysaccharide’s yield is 4.68 %.2. Extraction and purification of EAP, and explore its various physical and chemical properties. Use infrared scan EAP1-EAP4, we can know that, EAP1-EAP4 are all have αGlycoside bond in them. Use UV scan EAP1-EAP4, we can know that, EAP1-EAP4 are all been well purification. Use HPGPC to test EAP1-EAP4, know that the molecular weight were 48791.26 Da, 32646.78 Da, 19251.37 Da and 1197.34 Da.3.Antioxidant activity of EAP been explored. Determining EAP1-EAP4 radical DPPH? clear IC50 values were 0.29 mg/ml, 0.52mg/ml, 0.37mg/ml, 0.53mg/ml. Determining EAP1-EAP4 radical OH? clear IC50 values were 0.47 mg/ml, 0.66 mg/ml, 0.62 mg/ml and 0.73mg/ml.Overall, use single factor test and response surface methodology established the optimum extraction conditions of EAP isolated and purified by a uniform EAP obtained after different molecular weight, for the subsequent structure solution, structure-activity relationship studies provide a basis.
