After four and six weekstreatment and observe the experiment rats kidney weight and body weight;creatinine （Scr）,blood urea nitrogen （BUN）, phosphorus,calcium, blood red blood cells （RBC）, thehemoglobin （HGB）, aspartate transaminase （AST） and alanine aminotransferase （ALT）changes; Using HE dyeing observed the renal pathology change; immunohistochemistrymethods were used in detection kidney tissues TNF alpha and BMP-7expression. Useenzyme linked immunosorbent assay （Elisa） method for detection of serum TGF-in Beta1expression;use Protein imprinted （Western） detection of TGF-in Beta1, Smad-6expression inrenal tissue.Results: After treatment4weeks and six weeks all is model control group and normal controlgroup comparison: Scr, BUN were elevated significantly （P <0.01）, RBC, HGB werereduced significantly （P <0.05）, Kidney weight/body weight ratio increased, the blood Ca/P ratio decreased, AST, ALT no significant changes （P>0.05）, Renalhypertrophy,inflammatory cells within the organization and Kidney tissue fibrosis,TNF alphapositive expression rate increased significantly （P <0.05）; TGF-beta1expression in renaltissues and blood rate increased significantly （P<0.05）, Smad-6, BMP-7protein expression inrenal tissues significantly reduce （P<0.05）.Shenshuaining capsule group, every concentrationof upbear the clear and downbear the turbid capsule group, compared with the model group:Scr, BUN were significantly reduced （P <0.05）; Kidney weight/body weight ratio decreased, the blood Ca/P ratio increased, RBC, HGB was significantly elevated （P <0.05）;AST, ALT showed no significant change （P>0.05）; Renal hypertrophy inflammatory cellswithin the organization and Kidney tissue fibrosis were reduced; the TNF alpha positiveexpression rate in renal tissue was significantly reduced （P <0.05）, Blood and renal tissue ofTGF-β1expression rate was significantly lower （P <0.05）, in renal tissue of Smad-6andBMP-7protein expression was significantly increased （P <0.05）. upbear the clear anddownbear the turbid capsule in low-concentration group compared with Shenshuainingcapsule group no significant difference in indicators （P>0.05）, which mean and standarddeviation of the minimum; upbear the clear and downbear the turbid capsule everyconcentration group compared with each other no significant difference in indicators （P>0.05）,which mean and standard deviation of the minimum; the Treatment for six weeks, upbear theclear and downbear the turbid capsule in low concentration group the observed indicatorsmore than4weeks of treatment works well.Conclusions: Upbear the clear and downbear the turbid capsule can effectively control theoccurrence and development of chronic renal failure. Speculate upbear the clear anddownbear the turbid capsules mechanism of action may improve renal function, fibrosis,regulating Ca, P metabolism, ease of renal anemia, inhibition of renal hypertrophy. Upbearthe clear and downbear the turbid capsule may be restrain TNF alpha in the expression of thekidney tissues to reduce resistance of erythropoietin extend the red blood cells life, reduce theinflammatory cells infiltration, increased the red blood cells generate by cleared urine toxins,plays the role of the treatment of renal anemia. through increased Smad-6, BMP-7proteinexpression reduced TGF-β1protein expression, thus inhibition of TGFβ^-1 signal transductionpathway to the cell nucleus, while reducing renal interstitial fibrosis that one of themechanisms.
