Objective To investigating the expression of VEGF in the tissue of renal cell carcinoma(RCC) and in peripheral blood.And to investigating their correlation in differrent pathologicalgrades and clinical stages of RCC. Method1. The expression of VEGF in the tissue of32cases of RCC and10cases of normal kidney was examined by real-time fluorescent quantitativePCR (RFQ-PCR).2. Using RFQ-PCR examined the expression of VEGF in tissue nearby renalcell carcinoma of24cases,and compared with the expression in tissue of10cases of normalkidney.3.The expression of VEGF in peripheral blood of32cases of RCC and of30healthypeople was examined by the same method. Each group were analyzed by SPSS11.5forWindows software. Results1. VEGF protein was significantly expressed in the tissue of RCC.The△CTvalue in the RCC group was8.24±1.06, it was markedly higher than that of thenormal kidney group, while its△CTvalue was13.44±0.81;2. In the poorly differentiatedgroup,the differentiation group and the well-differentiated group, The△CTvalue of VEGF wasdifferently7.06±0.64,7.74±0.77,8.74±0.78. There was statistically significant difference indifferent pathological grades and clinical stages.(P <0.05); In T1aperiod group, The△CTvalueof VEGF was8.82±0.86,while in T1bperiod group,T2aperiod group,T2b period group and≥T3period group,the△CTvalue of VEGF was differently8.77±0.85,8.09±0.87,7.28±0.74and6.70±0.47. There was statistically significant difference in each group of different clinical stages,except T1agroup vs T1bgroup.(P <0.05).3. There was no statistically significant difference ofthe△CTvalue in tissue nearby RCC of T1/T2agroup (13.30±1.15/13.25±1.09) compared withnormal kidney tissue group(13.44±0.81),howere,in T2band≥T3groups,the△CTvalue11.58±2.55and8.69±0.47were significantly difference between with control group.4.The△CT value of VEGF in preoperative peripheral blood of32cases of RCC was7.31±0.95,while thevalue in healthy people group was7.90±0.70. There was statistically significant difference ineach clinical stages and pathological group. Conclusion1. The expression of VEGF wassignificantly higher in RCC than in normal kidney tissue,while There was no VEGF expressedin the tissue of normal kidney group.2. The expression of VEGF in RCC was positively relatedto the pathological grades,clinical stages.So dececting VEGF in peripheral blood of RCCpatients could be a new method for assistant diagnosis of RCC.we could evalue themalignancies of RCC.3. If RCC patients with≤10cm localized tumor underwented the partialnephrectomy, more than0.5cm from the tumor boundary, the tumor should be ableto completely removed.4.The expression of VEGF in peripheral blood of RCC patients wassignificantly higher than healthy people. Therefore, detecting the expression of VEGF inperipheral blood of RCC patients could play an important role in diagnose and the forecasting oftumor micro-metastasis and recurrence after operation of initial RCC.
