Aim To investigate the-344T/C and intron2conversion polymorphisms of aldosterone synthase gene (CYP11B2) for an association with stroke and hypertension in the North Chinese Han population.METHODS A case-control study was taken from332IS patients and250healthy controls matched for age and sex. Genotypes of-344T/C polymorphism was determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and the intron2conversion polymorphism was genotyped using two separated polymerase chain reactions. Logistic regression was used to adjust for confounding variables.RESULTS Compared to control group, IS group had higher frequencies of genotype TT (60.2%VS45.6%, P<0.01). T allele frequency in case group was significantly higher than those in control group (78%VS69.2%)(P<0.01). An association was found between TT genotype and ischemic stroke[odds ratio=1.572,95%CI (1.095-2.258), p=0.014]. However, we did not find any significant association of intron2polymorphism with stroke.TT genotype frequency in hypertension subgroup (54.8%) was significantly higher than that in control group (36.3%)(P<0.01). An association was found between TT genotype and hypertension [OR=1.90,95%CI1.131-3.193, P=0.015].CONCLUSION CYP11B2gene-344T/C polymorphism might be associated with ischemic stroke and hypertension in North Han Chinese polulation. Intron2polymorphism might not be associated with increased stroke and hypertension susceptibility.
