Objective: The effect and its potential mechansim ofbaicalein on the histological structure, apoptosis, the migration andinvasion of human hepatocellular carcinoma cell stain SMMC-7721tran-splanted tumors in nude mice were investigated, in order to provide exp-ermental evidence and new mode for baicalein in primary hepatic carcin-oma clinical treatment. Methods:1. BALB/C (n/n) nude mice were usedto replicated SMMC-7721tranplanted tumor model, and they wererandmonly divided into experimental and control groups when theirtransplanted tumors growed to8mm in dianmeter. Each nude mice ofexperimental group was treated with baicalein(20mg/kg/d)by intraperit-oneal injection, and the control group treated with DMSO by the sameway. After14days, nude mice were killed, measure the volume andweight of the transplanted tumors, the tumor inhibition ratio(IR) wascalculated by the formulation: IR=(1-transplanted tumor weight ofexperimental group/transplanted tumor weight of control group)×100%.2. The protein of Survivin and Ezrin expression in SMMC-7721transplanted tumor were detected by IHC.3. The mRNA of Survivin andEzrin expression in SMMC-7721transplanted tumor were analyzed by RT-PCR.4. The histological structure of SMMC-7721transplanted tumorwere observed by light microscope.5. All of the datas were analyzed bystatistical software SPSS16.0. Results: Both of volume and weight of theSMMC-7721transplanted tumors in the experimental group weresignificently decreased than that in the control group(P﹤0.05), thetumor IR was20.80%±10.27%.2. The histological structure ofSMMC-7721transplanted tumor were observed by light microscope, anumber of necrosis, were showed in the experimental group, in thecontrol group the SMMC-7721transplanted tumor cells were arranged innests and showed significant heteromorphism and the necrosis waslimited.3. The protein of Survivin and Ezrin expression in SMMC-7721transplanted tumor of experimental group were significantly decreasedcompared with the control group(P﹤0.05)4. The mRNA of Survivin andEzrin expression in SMMC-7721transplanted tumor of experimentalgroup were significantly down-regulated compared with the controlgroup(P﹤0.05). Conclusions:1. The baicalein may inhibit the growth ofSMMC-7721transplanted tumor in nude mice.2. The baicalein maydown-regulate the expression of protein and mRNA of Survivin and Ezrinin SMMC-7721transplanted tumor.3. Baicalein down-regulate theexpression of survivin and Ezrin to promote the apoptosis ofhepatocellular carcinoma cells and inhibit the metastasis and invasion ofhepatocellular carcinoma cells.4. Baicalein is expected to become the new high efficiency and low toxicity drugs for clinical treatment ofhepatocellular carcinoma.
