| BackgroundBreast cancer is one of the most common malignant tumor among women, invasionand metastasis are the main causes of death, so study the mechanism of invasion andmetastasis is importent to the treatment of breast cancer.ObjectiveTo investigate the expression of CD44v6and Fascin-1in human breast carcinomatissue and adjacent normal tissue,and show the relationship between them and clinicalpathological characteristics of breast cancer and reveal their significance in thedevelopment and progression of breast cancer, at the same time, explore the correlationbetween CD44v6and Fascin-1protein during the development of breast cancer, andscreen the suitable breast cancer cell lines and provide gene therapy for breast cancer withrelative reference.Materials and methods1. ImmunohistochemistryChoose breast tissue chip for specimens, among them selected65cases with perfectclinical data as immunohistochemical specimens, each specimen collection tissue adjacentto carcinoma and tumor tissue of each specimen, then select65cases normal breast tissueas control group, and analyze the expression level of CD44v5and Fascin-1in the specimen of each group. Divide these specimens into groups according to the clinical pathologicalcharacteristics (age, tumor size, histological grade, lymph node metastasis, clinical stage,TNM,ER and PR receptors, c-erbB-2,ki67,P53, P16, E-cadherin) of obtained specimen.Observe the differential expression of C4D4v6and Fascin-1in breast cancer tissue andinvestigate the correlation between them and basical clinical pathological feature of breastcancer, understand the meaning of CD44v6and Fascin-1in the development andprogression of breast cancer and analyze the relevance between them2. Western blot and RT-PCRBreast cancer cell line MCF-7,T-47D,MDA-MB231and normal breast tissue cellswere selected as tissue cell culture, analyze the expression levels of CD44v6and Fascin-1among cell lines by western blot, meanwhile, screen breast cancer cell line by RT-PCR andselect suitable cell lines for the next step of gene interference.Results1.The expression of CD44v6and Fascin-1in the65cases of breast cancer tissuesignificantly increased as compared with that of cancer adjacent normal tissue (P <0.05),the expression of Fascin-1was not obviously correlated with breast cancerpatients’age,grade,er,pr,cerb-2,ki-67,p53,p16(P>0.05), but related to tumorsize,TNM,lymph node metastasis (P <0.05); The expression of CD44v6protein was notsignificantly correlated with breast cancer patient age, tumor size, ER, PR, E-cadherin, P53,and P16(P>0.05), but relate with tumor staging, lymph node metastasis, Cerb-2, and Ki-67(P <0.05).2. The expression of CD44v6and Fascin-1in breast cancer cell detected by western blot ishigher than that in the control group,p<0.05,which were considered statistically significant,and the expression of Fascin-1in the group of mcf-7is higher than the other three groups,Cd44v6protein expressed in T-47d group was higher than other groups, the same resultwas confirmed by RT-PCR.Conclusion:Our result suggested that the differential expression of CD44v6and fascin-1exist inthe breast cancer tissue and adjacent normal tissue, and the over expression of CD44v6and fascin-1was closely associated with lymph node metastasis stage and grade in breast cancer,while there is no correlation between them in the development and progression of breastcancer metastasis, It also help us screen suitable cell for lines gene interference therapy inbreast cancer,which play an vital role in the diagnosis and treatment of breast cancer. |
