Objective: Study on cluster of differentiation147(CD147) and matrixmetalloproteinase-7(MMP-7) expression in cholangiocarcinoma.Reveal therelationship between the CD147and MMP-7expression in cholangiocarcinoma andvarious clinical pathological data, as well as the relation between CD147and MMP-7protein.Study of CD147and MMP-7in the mechanism of the occurrence and tumorcells in tumor growth effect.Analysis of the prospects in tumor diagnosis andtreatment of bile duct carcinoma.Methods: The experiments were collected in the First Affiliated Hospital ofNanchang University between2009January to2013June, bile duct cancer operationresection in44cases of paraffin embedded specimens, all specimens were confirmedby pathological diagnosis.In all patients were not receiving preoperative all kinds oftumor treatment.The control group specimens of30cases, bile duct cancer radicaloperation for resection of para tumor tissue specimens, and confirmed by pathologicdiagnosis without tumor cells.Using immunohistochemical(IHC), to detect theexpression of CD147and MMP-7protein in bile duct carcinomas and the adjacenttissue samples.Application of Image Pro Plus6image analysis software,immunohistochemistry staining intensity of expression.The use of SPSS statisticalsoftware l7.0system, detection methods: normality test, analysis of variance(ANOVA), LSD test, t test, linear correlation analysis, X2test, and the data werestatistically analyzed, P<0.05difference has statistics significance.Results: CD147and MMP-7expression in cholangiocarcinoma experimentalgroups, the mean values of PU were34.2±9.5,57.5±15.4.CD147and MMP-7inthe control group, the mean values of PU in cancer tissues were12.2±4.2,12±2.5,.Expression of CD147protein and MMP-7protein expression in cancer and bileduct cancer tissues compared group, significant differences in the expression ofcholangiocarcinoma (P<0.05).Using the linear correlation analysis confirmed that the expression of CD147was positively correlated with MMP-7in cholangiocarcinoma(r=0.784, P<0.01).CD147in cholangiocarcinoma is mainly expressed in the cell membrane,cytoplasm and nucleus no expression or low expression;CD147expression in tumorcompared with expression of high differentiation group and differentiation group andthe expression of low differentiation group, with statistical significance (P<0.05),expressed as tumor differentiation, PU value increased significantly, tumordifferentiation, PU value increased significantly. Further analysis showed that theexpression of CD147in tumor invasion depth, clinical stage of TNM, the differencewas statistically significant (P<0.05),But no significant correlation with patient age,gender, tumor location, neural invasion (P>0.05).The experimental results suggestthat the CD147may be associated with cholangiocarcinoma infiltration andprognosis.According to the clinical TNM stage, CD147expression in I, II and III, IVare compared with each other, with significant difference (P<0.05), expression ofCD147in tumor and tumor clinical TNM staging.MMP-7in cholangiocarcinoma cells were mainly located in the cytoplasm, cellmembrane and nuclear low or no expression.MMP-7expression in tumor comparedwith expression of high differentiation group and differentiation group and theexpression of low differentiation group, with statistical significance (P<0.05),expressed as tumor differentiation, PU value increased significantly. MMP-7proteinexpression in cholangiocarcinoma statistically significant comparison between TNM stagegroup (P <0.05),but no statistical significance with the patient’s age, gender, tumorlocation, neural invasion, tumor invasion depth correlation (P>0.05).Findings from44patients with bile duct carcinoma in the medical records,clinical TNM staging of cholangiocarcinoma is associated with the location of thetumor (P<0.05), but no statistical significance with gender, age, tumor differentiationcompared (P>0.05).Manifestations of hilar bile duct carcinoma staging than distalbile duct cancer later, gender, age, tumor location, tumor differentiation and depth ofinvasion, perineural invasion compared with no statistical significance (P>0.05).Conclusions:CD147and MMP-7in cholangiocarcinoma tissue expression maybe correlated with the tumor occurrence and development mechanism.CD147and MMP-7is associated with the biological behavior characteristics and prognosis oftumor.Combined detection of CD147and MMP-7help in the early diagnosis ofcholangiocarcinoma, treatment and prognosis. There was a positive correlationbetween CD147and MMP-7in cholangiocarcinoma tissues, but the mechanism andthe effect of presence of each other between CD147and MMP-7will be throughfurther research.
