Objective:to investigate the inhibitory action of curcumin for the subcutaneous transplantation of cancer of the liver of liver cancer,and the researchonthemolecularmechanismofthe anti-inflammatory-anti-hepatocellular carcinoma in the TLR4 pathway.Methods:1.42 rats were used in the establishment of the model of transplantation of liver cancer in liver cancer,and 40 were successfully made in 15d.Then 40 successful building of subcutaneous transplantation of liver cancer tumor nude mice completely random Ly divided into 4 groups,respectively HDC(High doses Curcumin)group(10),the LDC(Low doses Curcumin)group(10),the positive control group(10),model group(10),the other set to normal group(10),a total of five groups.When building 15d after began to fill the stomach,HDC group to the dose of 286.65 mg/kg/d(0.4 m L/d daily,concentration of 0.4 mg/m L),LDC group to the dose of57.33 mg/kg/d(0.4 m L/d daily,concentration of 0.4 mg/m L),continuous dosing14d;The positive control group,5-fu:50mg/kg/times+cisplatin:5mg/kg/kg/(concentration of 1.25 mg/m L+0.25 mg/m L)1 time/W,abdominal injection.The normal group,the model group gave an equal amount of 0.4 m L/d,and 0.9%of the sodium chloride solution to the stomach,the continuous medicine 14d.2.After 14 d drug intervention to be put to death,observe the general condition,tumor rate of nude mice and subcutaneous transplantation tumor growth,subcutaneous transplantation tumor,weighing tumor weight,calculate the inhibitory rate.The Western blot method detects the positive expression of TLR4(toll-like receptor 4),NF-_?B(nf-kappab),and i NOS(Inducible nitric oxide synthase)in the transplanted tumor tissue.Results:1.ofcurcuminonlivercancernudemicesubcutaneously transplantedtumorhassignificantinhibitoryeffect,HDCgroup(364.64±42.65 mm3),the LDC group(547.97±43.12 mm3),positive control group(646.76±168.66 mm3)the average tumor weight and subcutaneous transplantation of liver cancer tumor(1285.97±79.38 mm3)compared with model group were lower(p<0.01),which the average tumor weight minimum HDC group,and the inhibitory rate is highest;2.Western blot results showed that hepatocellular carcinoma in nude mice subcutaneous transplantation tumor tissue TLR4 protein positive expression:HDC(0.116±0.028),the LDC group(0.24±0.037),significantly in positive control group(0.27±0.025)were lower than that of model group(0.37±0.04)(p<0.01);The HDC group was significantly lower than the positive control group(p<0.01).Compared to the LDC group(p<0.01)compared to the LDC group(P<0.01),the LDC was significantly lower than the positive control group(P<0.01).3.The subcutaneous transplanted tumor tissue of liver cancer in NF-_?B protein positive expression:HDC(0.12±0.04),the LDC group(0.16±0.035),positive control group(0.22±0.021)were lower than that of model group obviously(0.37±0.03)(P<0.01);The HDC group was significantly lower than the positive control group(P<0.01).Compared to the LDC group(P<0.01)compared to the LDC group(P<0.01),the LDC was significantly lower than the positive control group(P<0.01).4.INOS protein positive expression in liver subcutaneous transplantation tumor tissue:HDC group(0.14±0.033),the LDC(0.24±0.034),positive control group(0.35±0.045)were lower than that of model group(0.47±0.043),(P<0.01),of which the HDC group compared with the positive control group,significantly lower than the positive control group(P<0.01);Compared to the LDC group(P<0.01)compared to the LDC group(P<0.01),the LDC was significantly lower than the positive control group(P<0.01).Conclusion:1.Curcumin has a significant inhibitory effect on the growth of the tumor growth in the skin of the hypodermic carcinoma of the liver,and is associated with the dose.Curcumin inhibits the release of NF-_?B and i NOS by inhibiting the TLR4and inhibits inflammation and ACTS as an anti-liver cancer.
