Salvia officinalis is a genus of Salvia Lamiaceae with a wide range of physiological activity. Modern pharmacological experiments show that Salvia officinalis has a lot of benefits, such as expanding the coronary artery, improving microcirculation and protecting heart, it can also inhibit platelet from coagulating and aggregating. At the same time, it may improve the ability of the body anti-hypoxic, anti-tumor and anti-virus. In a word, it is one kind of the important medicinal plants. In this paper, the extraction of Salvia officinalis oil, its purification and its antioxidant activity were focused. Through research, new functions and uses of this essential oil may be discovered and developed.(1)Two methods including Supercritical CO2extraction and Steam distillation extraction were used to extract essential oil. Salvia officinalis was choose as the raw material and confirmed the optimal extracting conditions of two extraction methods by using the extraction rate as index. The following conditions of Supercritical CO2extraction were optimal:The granularity of Salvia officinalis is between40and60mesh, temperature of extraction is50℃, the pressure is15MPa, the flow rate of carbon dioxide is20L/h, time of extraction is120min, and temperature of separation is40℃. The rate of Salvia officinalis oil obtained by Supercritical CO2extraction was6.01±0.13%. The optimal extracting conditions of Steam distillation extraction were as following:the granularity of our material is between40and60mesh, time of distillation is1.5h, solid-liquid ratio is1:8, soaking time is1.5h, the consistence of NaCl as distilling additives is2%(w/w), microwave-assisted extraction time is30s. The Salvia officinalis essential oil extracted by Steam distillation extraction rate is3.04±0.15%.(2) Through determining physical and chemical properties and the component content of Salvia officinalis essential oil extracted by Supercritical CO2extraction and Steam distillation extraction, it can be seen the essential oil extracted by Steam distillation is close to Spanish Salvia lavandulifolia Vahl in physical and chemical properties, but the component contents of them are quite different from each other; Several indicators of Salvia officinalis essential extracted by supercritical extraction are slightly beyond the standard of it. (3) By Studying on the Salvia officinalis essential oil extracted by supercritical CO2extraction and Steam distillation methods and comparing their antioxidant activities in vitro, the Salvia officinalis essential oil extracted by both method has strong antioxidant capacity examined by DPPH, hydroxyl radicals, super oxide radicals and it will increase with dose. The Salvia officina/is essential oil extracted by supercritical CO2extraction has stronger antioxidant capacity than extraction by Steam distillation in this three oxidation system.(4) Silica gel column chromatography was used to purify the Salvia officinalis essential oil extracted by supercritical CO2extraction. The results showed when eluent ratio was petroleum ether:ethyl acetate=9:1(v/v), purification worked best. After GC/MS analysis, the main component of Salvia officinalis essential oils were enriched, the total content of α-pinene, α-thujone camphor, linalool, linalyl acetate and caryophyllene were enriched by8.5%to23.3%.
