ObjectiveTo observe the pharmacological effect of ginsenoside Rb1on blood pressure, the balance ofTh17/Treg cells and oxidative stress index in spontaneously hypertension rats, and to explorethe possible mechanism.Methods3612-week-old male SHRs were randomly divided into four groups, large doses ofginsenoside Rb1and small dose group and blank group (saline), positive control group(amlodipine).9normotensive Wistar Kyoto rats (WKY) only for the normal control group(WKY group). A tail cuff method was carried out to determine the arterial blood pressure at thetreatment of0week and the end of12weeks respectively. Blood was also collected to analysisTh17and Treg cells ratio by flowcytometry. the related cytokines IL-17, IL-10as well asIndicators of oxidative stress such as SOD activity,MDA level and ox-LDL were detected byElisa at the same time. At the end of12weeks, animals were sacrificed after aortic blood iscollected, and then immune cells which infiltrate vessel wall were observed usinghistochemical methods.ResultAfter12weeks treatment with ginsenoside Rb1, the blood pressure of large and small dosegroups were significantly decreased than pre-treatment (high dose group decreased by19.25%,and low dose by7.91%), but not superior to the amlodipine group (decreased by24.17%);When Rb1of high dose group and amlodipine group compared with the control grouprespectively, the difference was statistically significant(P<0.05). Ginsenoside Rb1high andlow dose groups can both decrease the proportion of Th17cells and increase the proportion ofTreg cells. The regulation of Treg is more significant (high dose group increased2.40times,low dose increases1.49times). However, effect of amlodipine group on the distribution of Th17/Treg cell and the ratio is not obvious. Accordingly, the IL-17levels which correspondingto the Th17activity were down regulated and the IL-10which corresponding to the activity ofTreg increased1.7times after Rb1treatment. Immunohistochemical analysis showed thatdecreased Th17and upregulated Treg were shown in the vascular wall in Rb1treated groups.Detection of oxidative stress index found,after Rb1treatment, MDA, SOD and ox-LDL levelsall have a certain degree of improvement, especially for the MDA and ox-LDL changed mostsignificantly, in the high dose group were down2.41times and1.83times respectively,compared with the saline group,respectively P<0.01and P<0.05, the differences were verystatistically significant, showing a strong antioxidant activity of Rb1.ConclusionGinsenoside Rb1can significantly reduce blood pressure as well as adjust the immune systemby changing the Th17/Treg cell homeostasis in spontaneously hypertension rats. In addition, Itshowed significant antioxidant effect of ginsenoside Rb1which predict a further developmentand use in clinical.
