Euphorbia ebracteolata Hayata, distributed in Anhui, Henan, Jiangsu, Shandong,Hubei, Guangdong, Guangxi Provinces, has been used for the treatment of edema,abdominaldistension, scrofula and ringworm. This thesis is focused on the chemicalinvestigation and antigout activity research of the extract of E. ebracteolata.1. Chemical constituents of Euphorbia ebracteolataIn order to search bioactive substances from E. ebracteolata, systematicalchemical investigation of the speciesis was carried out.36compounds were obtainedamong which19compounds were identified by chemical and spectroscopic mothord.They are14β-hydroxy-11-oxo-ent-abieta-8(9),13(15)-dien-12α,16-olide (EE-14),8β,14β-epoxy-13,15-abiatene-12,16-olide (EE-19)、8β,14α-dihydroxy-13,15-abiatene-12α,16-olide(EE-20)、8β,11α,14α-trihydroxy-13,15-abiatene-12α,16-olide (EE-23), EbracteolatainC(EE-33), Jolkinolide E(EE-03),2,4-dihydroxy-3-methyl-6-methoxy-acetophenone(EE-01),β-amyrin acetate(EE-02),2,4-dihydroxy-6-methoxy-3-methyl-acetophenone(EE-04),2,2’,4,4’-Tetrahydroxy-6,6’-dimethoxy-3,3’-dimethyl-7,5’-bisacetophenone(EE-05), Jolkinolide A(EE-06),β-sitosterol(EE-8), cycloart-23-ene-3β,25-diol(EE-9),3,3’-diacetyl-4,4’-dimethoxy-2,2’,6,6’-tetrahydroxydiphenylmethane(EE-10),24-ethyl-5,22-cholestadien-3-betaol (EE-11), Ent-atisane-3α,16β,17-triol(EE-17),Ent-kaurane-3-oxo-16α,17-diol (EE-25), Scopoletion(EE-29),Ent-11α-hydroxyabieta-8(14),13(15)-dien-16,12α-olide(EE-30). Among them,EE-14,EE-19,EE-20,EE-23and EE-33are new compounds. EE-3and EE-17wereisolated for the first time from this species.2. Antigous activity investigation of Euphorbia ebracteolataA preliminary study ofEuphorbia ebracteolatabiological activity was carried out.Oxygen potassium cyanatewas evolved to induc KM mice as Hypouricemic model.The influence of Euphorbia ebracteolataethanolextractsto uric acid levels inurine, serumand liver in mice were shown. The effects of Euphorbiaebracteolataethanolextracts on hepatic XOD activity were also investgated.Theresultsindicatedthat high dosegroup induced hypouricemic efficacy to reduce uricacid production and enhancing urate excretion in hyperuricemic mice by suppressingliver XOD activity. The pathological changes in the kidney of mice wereobservedthrough microscope.In the model group mice,kidney cells changesignificantly: loss of normal structure, degenerationof part of the renal tubularepithelial cells in vacuoles, obscission of part of the renal tubular epithelial cells. Themodel groupshowed significant differences compared with the normal group.Positivesilymarin group essentially showed normal kidney cells without necrosis. High dosegroup induced inflammation in mice and formation of a proteinuria. It inferred thatEuphorbia ebracteolatahas a treatment activity of hyperuricemia and gout.
