BackgroundSpinal cord injury is severe trauma of the central nervous system.the recoverycapability of feeling and motor function are very limited after spinal cord injury,and the morbidity is very high. But, there is no effective clinically method for itsintervention and treatment. Studies have shown that spinal cord function disordersare decided by Primary and secondary damage.the primary damage is oftenunavoidable,while secondary damage can be Partially reversed by aggressivetreatment,which Provides the opportunity for a variety of Possible treatments.Recently many studies are in the Process of using drugs to interfere with thissecondary damage,which expects to mitigate secondary damage, eliminatedisincentives nerve regeneration and restorate damaged nerve function by improvingthe microenvironment of the site of injury.Desuetude found that Hepatocyte GrowthFactor(HGF) is a cytokine with multiple biological activities.It Plays the role ofendotrophic in various types of nerve cells. It can Promote angiogenesis and improvemicrocirculation. Also it can also reduce the formation of astrocytes and Promoteaxonal growth.But there are few reports have shown whether HGF can Promoterecovery of spinal cord injury in rats electromotor function.ObjectiveThis subject use modified Alien’s method to build rat spinal cord injury model.to inject hepatocyte growth factor through subarachnoid catheter. Investigatethe effect of HGF on electromotor function recovery by detecting SEP and BBBrating changes of each group at different time after spinal cord injury. Observing theeffects of SCI astrocytes and nerve cells by comparing of HGF after spinal cordinjury with GFAP by immunofluorescence staining, which we can explore thePartially mechanism of HGF recovery function after spinal cord injury.Method1.24female SD rats divided into control group and experimental group,usingmodified Alien’s method build model and stay tubes in subarachnoid to administratethe rats;2.Inject HGF and equal PBS through the catheter respectively immediately andafter the injury once a day for7days,then we remove the catheter.3. Evaluating neurological recovery of motor function in the Preoperative andpostoperative7,14,21,28days by SEP and BBB rating.4. Sacrificing the rats on7th,28thday, then comparing changes in the structure oftwo groups after spinal cord injury by hematoxylin-eosin (HE) andimmunofluorescence staining.Results1. Build a unified standard model of spinal cord injury and administration issmoothly for one week.2. Detection of SEP latency: the incubation Period before injury were23.11±1.19ms, the incubation Period after injury became longer obviously, but theincubation Period at various Points in the treatment group were significantly shorterthan the control group(P<0.05).the results suggest that HGF may Promote therecovery of neurological function SCI in rats.3. BBB rating:the rating before injury was21, after injury appeared as completely Paralyzed was0, then there are different degrees of recovery, there wasno significant difference in the two groups at7th day after injury,but the recoveryafter injury of treatment group at other time was significantly higher than controlgroup(P<0.05).the results support the conclusions of somatosensory evokedPotentials.the results suggest that HGF may promote the recovery of neurologicalfunction SCI in rats.4.HE and GFAP shows there are a large number of increased cell body andProjections extending astrocytes in the area of spinal cord injury; But organizationalstructure in the treatment group is better than the control group, And HE stainingshowed that the maximum diameter of the damage zone and necrotic area treatmentgroup than the control group, and a statistically significant(P<0.05). Furthersuggest HGF can Promote recovery of neurological function after SCI in rat.ConclusionHGF can Promote to restore nerve function in rats with spinal cord injury...
