Objective:Through the construction of gastric cancer cells transplantation tumor in nude mouse model, and implanted the Iodine-125particles, to observe the growth and the proliferation of tumor, draw the graph of tumor growth, measure the weight and calculate the inhibition rate of tumor, then detected the expression of gene and protein of the nuclear factor kappa B in tumor tissue,to clarify the effect of the Iodine-125particles on the expression of nuclear factor kappa B in gastric cancer cells.Methods:In human gastric cancer cell line SGC-7901in BLAB/c-/nu tumor bearing nude mice as animal model, and are randomly divided into three groups (n=30).One is blank controrl group, which to be implanted empty seeds, without Iodine-125radioelement(OMBq); The second is experiment group, which to be implanted Iodine-125seeds (0.6mCi). The third group is a control group,which is nothing implantation. Every3days to measure the size of tumor,calculate the volume and draw the graph of tumor growth. When implanted Iodine-125seeds28days, Put to death all nude mice and obtain tumor specimens. Compare the inhibition rate of the experimental group and control group and blank control group. RT-PCR method was used to detect the expression of the nuclear factor kappa B’s gene in tumor tissue, and with the expression of Western blot method in tumor tissue for nuclear factor kappa B’s protein.Results:1. Experimental group on the inhibitory rate of the blank control group was43%, experimental group on the inhibitory rate of the control group was38%. 2. Messenger RNA and protein expression of nuclear factor kappa B’s gene in Iodine-125particles implantation in the experimental group were significantly lower than that of the control group, the experimental group compared with control group and blank control group, the difference has statistical significance(P<0.05).Conclusions:1.Iodine-125particles can inhibited gastric cancer tumor growth significantly.2. This experiment confirmed that Iodine-125particles radiation on the effectiveness of human gastric tumor treatment of mice tumor model, and explained that Iodine-125particles after radiation therapy can inhibit the expression of the nuclear factor kappa B in tumor tissue.3. Iodine-125particles make mice inhibited the growth of gastric cancer,which is related to inhibit the expression of nuclear factor kappa B, these may be one of the molecular biology mechanism of the treatment of tumor occurrence and development.
