ObjectiveTo investigate the association between the serum level of MMP-9, the serum level of TIMP-1, MMP-9/TIMP-1and coronary artery ectasia (CAE). To study the distribution of single nucleotide polymorphism C/T in the promoter of MMP-9gene about CAE. To explore whether MMP-9gene polymorphism is related to coronary artery ectasia. To verify the correlation with the genotype distribution and the serum MMP-9level.Methods1. The clinical data of175consecutive patients were collected. These patients were divided into three groups according to coronary artery angiography(59patients with CAE,61patients with CAD and55patients with angiographically normal coronary controls). To evaluate the characters of clinical manifestation and coronary artery angiography of CAE.2. MMP-9, TIMP-1levels in all subjects included in this study were measured by enzyme linked immunsorbent assay (ELISA) method.3. The genotypes of MMP-9were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) methods in three groups.To analyze the MMP-9gene promoter-1562C/T genotype of three groups patients.Result1. The differnce was not statistically significant in three groups with the characters of clinical manifestation (P>0.05).2. The MMP-9level and MMP-9/TIMP-1were significantly higher between CAE group and NCA group(P<0.05); although the MMP-9level and MMP-9/TIMP-lin CAE group were higher than CAD group, it did not reach statistical significance (P>0.05). There were no significant difference of TIMP-1level among three groups(P>0.05).3. There were no significant difference of MMP-9level among the different types of CAE and among the different number of dilated coronary arteries(P>0.05).4. The genotype frequencies of CC, CT and TT were0.81,0.17and0.03in CAE group; the distribution frequencies were0.79,0.19and0.03in CAD group,the distribution frequencies were0.69,0.29and0.02in NCA group;but the difference was not statistically significant (x2=2.11,P=0.35). The allele frequencies of C and T were0.88and0.12in CAE group,0.87and0.13in CAD group,0.87and0.13in NCA group,The difference was not statistically significant in three groups (x2=0.04, P=0.98).5. There were no significant difference of the genotype frequencies among the different types of CAE and there were no significant difference of the genotype frequencies among the different number of dilated coronary arteries (P>0.05).6. There was no significant difference in MMP-9level in patients with T allele among three groups (P>0.05).Conclusion1.The serum level of MMP-9and MMP-9/TIMP-1were associated with the development of CAE.2. In CAE group, MMP-9gene promoter had different distribution of genotypes. But the frequencies of the genotype was significantly different from CAD and NCA.3. MMP-9gene polymorphism-1562C/T was not significantly correlated with CAE.T allele of MMP-9cannot be a genetic factor that may contribute to individual susceptibility for CAE.
