ObjectiveBy detecting serum YKL-40level and its relevant gene CHI3L1-329G/A (rs10399931polymorphism with coronary atherosclerotic heart disease (CAD) patients, coronary artery ectasia (CAE) patients and normal coronary artery (NCA), to explore the possible pathogenesis of coronary artery ectasia, So as to pursue possible way to diagnosis and treatment of coronary artery expansion.MethodsStudy population included175individuals who underwent coronary angiography in Tianjin first central hospital during Sep2011and Oct2013.Fifty-nine patients with isolated CAE,61patients with CAD,55age-and gender-matched control participants with normal coronary arteries (NCA),were included in the study.All the patients’clinical data was recorded as follows, such as gender, age, smoking, triglyceride, total cholesterol, low-density lipoprotein cholesterol, high density lipoprotein cholesterol, fasting plasma glucose, etc. Serum YKL-40levels were measured by enzyme linked immunsorbent assay (ELISA) method. The genotypes of CHI3L1-329G/A were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) methods. Statistic method:Analyze the data used the statistic software SPSS17.0. Chi square test were used to compare numeration data between two groups. Scale data were expressed with mean±sd, followed T test was used to compare scale data between groups. P<0.05was considered to be statistic significance.Results1、Serum YKL-40levels were significantly different among study groups: CAD:69.30±11.24ng/ml, CAE:70.50±10.96ng/ml, NCA:50.21±12.02ng/ml, CAE group and CAD group had significantly higher YKL-40levels than NCA group (P=0.000), there were not significantly different between CAE group and CAD group (P=0.720)2、Logistic regression analysis revealed only YKL-40level as the determinant of CAE (P=0.005). 3、The CHI3L1gene-329G/A genotype was in Hardy-Weinberg.4、Three genotypes, CC, CT, and TT, existed in rs10399931and there were no significant differences found in either the genotypic or allelic frequencies among study groups.5、The expression of CHI3L1gene-329G/A is not associated with serum YKL-40levels and CAE.Conclusion1、Serum YKL-40levels were found significantly high in patients with isolated CAE, which may participate in the occurrence and development of coronary artery expansion.2、Three genotypes, CC, CT, and TT, existed in rs10399931and there were no significant differences found in either the genotypic or allelic frequencies among study groups. CHI3L1gene-329G/A polymorphism may not contribute to individual susceptibility for CAE.3、There were no significant differences found in serum YKL-40levels among CC, CT, and TT three genotypes. CHI3L1gene-329G/A polymorphism were not related to serum YKL-40levels.
