ObjectivePithecellobium clypearia(Jaek)(Fabaeeae),a herbal drug, belonged to Pithecellobiium Mart genus, Leguminosae, which mainly are shrubs. The common name of P. clypearia called weixian tree. In the folk, also called popi tree, the heart tree, jiao long mu, ji xin tree, luo di jin qian, etc. They manly grow in zhejiang, fujian, Taiwan, guangdong, guangxi, yunnan. They are bitter and cold, have good effection of clear heat purging fire, anti-inflammation, detumescence, detoxification, dispel dampness and lian chuang. They are precious Chinese medicine and commonly used in guangdong. Now they have been developed into a variety of dosage forms of antibacterial antiphlogistic medicine preparations, such as hou er huan anti-inflammatory capsule, hou er huan anti-inflammatory particle, hou er huan anti-inflammatory tablet, which are widely used on clinical to treat children with hand-foot-mouth disease, acute respiratory infections, pneumonia, acute pharyngitis, acute enteritis and tonsillitis in children. To further comprehensive knowledge and understanding the chemical chemistry of the P. clypearia, explore the active ingredient of the plant pharmacology to ensure effective, safe, and more fully rational use the P. clypearia herbs, explore more of its pharmacological effects, used to treat more diseases clinically, this subject were determined based on the chemical constituents of the main ingredients again.Methods1. Using the column chromatography technology of silica gel and macroporous resin, ODS, SephadexLH-20, MCI, separated and purified the chemical composition of the methanol extract in ethyl acetate parts of the P. clypearia herbs, and11compounds were isolated from it. Through the modern spectral analysis methods (NMR, UV, IR, MS), reference substance contrast and comparison method, the structure of9compounds were identified. 2. The study of chemical composition to the P. clypearia found that the ethyl acetate part mainly contain flavonoids ingredients. This article used the high performance liquid chromatograph of yilite EC2000which producted by dalian to determinate the conten of the myricitin-3-0-α-L-rhamnpyranoside,5,3’,4,5’-tetrahydroxyflavan-7-gallate, quercitin-3-0-α-L-rhamnpyranoside in the herbs, concrete and perparations of the P. clypearia by gradient elution. The conditions of the mobile phase is organic phase (methanol, acetonitrile=70:30)-water (0.2%phosphoric acid)Results1. Separated the9compounds:gallie acid(4000mg), ethyl gallate(100mg), kaempferol-3-0-α-L-rhamnoside(100mg), quercitin-3-0-α-L-rhamnpyranoside (1400mg), querceti(250mg),5,3’,4’,5’-tetrahydroxyflavan-7-gallate(2500mg), luteolin(36mg),myricitin-3-0-α-L-rhamnpyranoside(42mg), the mixture of gallocatechin-7,3’-gallate and gallocatechin-7,4’-gallate (2060mg).2. The results of the simultaneous determination using HPLC methods shows that the conten of myricitin-3-0-α-L-rhamnpyranoside,5,3’,4’,5’-tetrahy-droxyflavan-7-gallate and quercitin-3-0-α-L-rhamnpyranoside in the herbs of the P. clypearia were0.36~2.23mg/g,6.46g-22.41mg/g,1.06-4.59mg/g; The content in the concrete were0.50mg/g,5.87mg/g,2.42mg/g; Content in the tablet were0.78~0.87mg/g,5.75~6.22mg/g,1.93~2.25mg/g; The content in the particles were0.86mg/g,3.81mg/g,1.24mg/g; The highest content in the capsule were1.3mg/g,6.18mg/g,2.90mg/g, The13sample content lowest, the content of5,3’,4’,5’-tetrahydroxyflavan-7-gallate was2.72mg/g, almost no myricitin-3-0-α-L-rhamnpyranoside and quercitin-3-0-α-L-rhamnpyranoside.CouclusionThe determination results showed that the content of myricitin-3-0-α-L-rhamnpyranoside,5,3’,4’,5’-tetrahydroxyflavan-7-gallate and quercitin-3-0-α-L-rhamnpyranoside in medicinal herbs which growed in different regions had vary widely difference. suggesting that the cause large differences in levels may be due to different environmental climate and soil differences; also found that different medicinal ingredient and content of the site also contained a larger difference, three flavonoid contents were higher in leaves, followed by the twig, the lowest levels in the stem or no. So, in order to ensure the quality of medicine, recommended standardized planting, the site should also be clear medicine. Three compounds are also differences in the content of different forms, but the samemanufacturer in the same do-sage form content or less. So recommended that appropriate dosage forms and the dose should be different depending on the treatment purpose and the differen treatment groups.Determination of the results can also be based on that of the HPLC method is simple, feasible and reproducible, sample preparation is not complicated, can be used for quality control and determination of the P. clypearia(Jaek) Benth.
