Astragalus membranaceus, a perennial herb, is one of the traditional Chinese tonics. The studies on chemical constituents and pharmacological parts of Astragalus membranaceus are the root. The whole plant is abandoned, which resulted in a tremendous waste of resources and destroyed the renewability of the resources. Therefore this paper studied the chemical composition of the leaves of Astragalus membranaceus Hance systematically and effect relationship and quality control of large components flavonoids.1. The leaves of Astragalus membranaceus Hance extract compounds have been Layer of n-butanol extract extraction and separation of the system and chemical structure identification by silica gel, ODS, Sephadex LH-20column chromatography and HPLC preparation chromatography. As a result,22compounds were separated by silica gel, ODS column chromatography and HPLC and the chemical structures of20compounds were fully determined by1D and2D NMR such as (24E)-ene-3B,6a,16B,27-tetraol-6a-acetyl-16B-hydroxy-9,19-cycloartane-3-O-B-D-xylopyranoside-27-O-B-D-glucopyranoside;(20R,24S)-11a,16B,25-trihydroxy-20,24-epoxy-9B,19-cycloartane-3,6-dione; soyasapogenol E;3B,21a,24-trihydroxyolean-12-ene;3B,22B,24,29-tetrahy droxyolean-12-ene; soyasapogenol B; pisumsaponin I; soyasapogenol B-3-O-B-D-glucopyranosyl(1->2) B-D-glucopyranoside (1->4)-a-L-rhamnopyranosyl; huangqiyiesaponin C; astragaloside II;3,3’,41,5,7-pentahydroxyflavone; quercetin-3-O-B-D-glucopyranoside; rhamnocitin-3-O-B-neohesperidoside;kaempferol-3-O-[6"-O-(trans-p-coumaroyl)]-B-D-glucopyranoside;rhamnociti n-3-O-B-D-glucopyranoside;rhamnocitin-3-O-B-D-glucopyranosyl-(1"’-2")-B-D-apiofurano slide; complanatuside; dehydrodiconiferyl-alcohol-4-B-D-glucoside;{22E,20S;24R)-5a,8a-epidioxy-ergosta-6,22-dien-3B-ol;(2S,3S,4R,8E/Z)-2-(2’-Hydroxylignoceroyl amino)-8-octadecene-1,3,4-triol-O-B-D-glucopyranoside. Among them there are10triterpenes and triterpenoid glycosides,7flavonoids and flavonoidal glycosides. The compounds of1,2were firstly reported, The compound named huangqiyenin L, huangqiyegenin IX, compounds of3,5,7,8,13,14,16,18,19,20was firstly found in the Radix Astragali and compounds of4,10,11,12,17was first assigned to the aerial parts of Astragalus.2. Determination of four kinds of flavonoids of Astragalus membranaceus leaves for the first time with the HPLC method from the leaves of Astragalus membranaceus. Methods:The separation was carried out on a Diamonsil-C18column (5μm,4.6mmx250mm). The gradient elution was employed using solvent A (methanol) and solvent B (water) at25℃; the gradient program was used as follows:0-50min, linear change from A/B (10:90, v/v) to A/B (85:15, v/v). The flow rate was set at1.OmL-min-1and the detection wavelength was at267nm. Results:All calibration curves of the four complanatuside(17), rhamnocitin-3-O-β-neohesperidoside(13); kaempferol-3-O-[6"-O-(trans-ρ-coumaroyl)]-β-D-glucopyranoside(14); rhamnocitin-3-O-β-D-glucopyranoside(15) indicated good linear regression within the test ranges, which showed between1.2960~20.736μg(r=0.9999)、1.760-28.16μg(r=0.9999)、0.6900-11.04ug(r=0.9996)、2.0660-33.056μg(r=0.9998), respectively. Also, the average recovery rates in the range of97.21%to102.4%and the RSD between1.7%and2.5%were able to reveal the good reliability of the method. The experimental results show that the quality evaluation method is accurate, simple and reproducible, and can be used for quality evaluation of flavonoids in the leaves of astragalus.This studyis not only to clarify thematerial basis for the efficacyof Astragalus the commonly used traditional Chinese medicine, rich in natural organic chemistry,provides essential scientific basis of independent intellectual property rights of Astragalus Ye Xinformulation is developed with Astragalus membranaceus leaves; and to establish the internationally recognized quality standards, improve the quality of Radix Astragali,guarantee and products, has the vital significance the development of natural resources of medicinal plants of new; two developed through non medicinal parts of Radix Astragali, will expand the medicinal sources, provides the possibility for the full utilization of the medicinal plantresources.
