Objective:Bi-directional solid state fermentation was a fermentation method which use medicinal fungi monascus as its fermentation strain, use ginseng powder as its medicinal fermentation medium, and constitute fermentative combination together. To investigate the technological conditions of Monascus-Ginseng bi-directional solid state fermentation by using Monacolin K as index, Then, the use of modern analytical techniques to prove fermentation products both containing medicinal composition and efficacy of Monascus and Ginseng. Ginsenoside transformation produce rare ginsenoside. Finally, through in vitro and in vivo pharmacology experiments investigated the reducing blood fat and the anti-tumor in vitro pharmacological activity of fermentation product.Methods:Single-factor test was adopted by the effects of pH, initial water content, culture time, inoculum size, filling quantity, age of seed, culture temperature, carbon source and nitrogen source on Monacolin K yield. And on this basis, orthogonal testing design was adopted by pH, choosing initial water content, culture time and inoculum size four factors as index. It is concluded that the optimal fermentation conditions by using Monacolin K as index. By using TLC and HPLC methods for qualitative analysis is the main active ingredients of fermented product. Finally, to study the reducing blood fatang and the anti-tumor in vitro pharmacological activity of fermentation product. Through special feed fed, set up the experimental hyperlipemia model in mice. Three doses, low(400mg/kg), medial(800mg/kg), high(1600mg/kg) doses. Affect the determination of fermentation product in mice serum TC, TG, HDL-C, LDL-C content. Fermented product in DMSO mixture200μg/ml,100μg/ml,50μg/ml,25μg/ml four concentration. To investigate inhibition rate of Hela by MTT.Results:The optimum conditions of Monascus-Ginseng bi-directional solid state fermentation as follows:100ml fermentation bottle containing10g ginseng powder, add4.5%soluble starch,1.2%yeast extract,0.2%NaNO3,0.2%KH2PO4,0.2%MgSO4,60%of initial water content, inoculate10%seed liquid of48h age, use lactic acid pH=2.0,26℃of culture temperature,15d of culture time. By the TLC method can be concluded that Monascus-Ginseng fermented product contain acid type Monacolin K. By the TLC and HPLC method can be concluded that a slight change in composition of ginsenosides in Monascus-Ginseng fermentation product, Monascus has certain ability of transformed ginsenosides, and transformed the rare ginseng saponin Rg3. In the experiment of reducing blood fat, compared with the experimental hyperlipemia model in mice, the content of TG, TC and LDL-C in mice administered medial doses, high doses and positive control group decreased sinificantly(0.01<p<0.05) or most sinificantly(p<0.01), the group administered medial doses and high doses showed a better effect than the positive control group. However this difference is not significant(p>0.05). The content of HDL-C in mice administered medial doses, high doses and positive control group increased most sinificantly(p<0.01), the content of HDL-C in mice administered high doses increased most sinificantly(0.01<p<0.05) than positive control group.In the experiment of anti-tumor, Monascus-Ginseng fermentation products has inhibitory effect on the growth of Hela. With the increase of drug concentration, the Hela cell inhibition rate is gradually increased. The different drug concentration were200μg/ml,100μg/ml,50μg/ml,25μg/ml. The corresponding inhibition ratio of Hela cell were50.89%,36.19%,23.18%,20.66%respectively. Compared with the25μg/ml group, the inhibition ratio of200μg/ml and100μg/ml group increased most sinificantly(p<0.01). The50μg/ml group showed a better effect than25μg/ml group. However this difference is not significant(p>0.05).Conclusion:1.Monascus can grow well in the ginseng fermentation substrate, and fermentation products both containing the medicinal composition and effect of Monascus and Ginseng.2.Fermentation products containing the Monacolin K of hypolipidemic active ingredient.3.Monascus has certain ability of transformed ginsenosides, and transformed the rare ginseng saponin Rg3.4.Monascus-Ginseng fermentation products has a good reducing blood fat efficacy.5.Monascus-Ginseng fermentation products has inhibitory effect on the growth of Hela.
