Gentiana scabra Bunge., bitter in taste and cold-natured, is a member of the Gentianaceae family and belongs to Gentiana. The plant has the effects of purging liver and gallbladder, clearing heat and dissolving dampness, analgesic, can treat the heat jaundice, genitals swollen itch, vaginal itching discharge, eczema, acute conjunctivitis, tinnitus and deafness, convulsions and other diseases. Studies have shown that G. scabra Bunge. has a variety of pharmacological effects, such as pr-otecting liver, gallbladder and stomach, diuresis, anti-inflamma tory, anti-allergy, anti-bacterial, anti-pathogens and so on. G. scabra Bunge., authentic ingredients of Changbai Mountain, is rich resources in Jilin Province. In order to further the development and use of authentic ingredients of Changbai Mountain, this study investigated the chemical constituents isolated from the root of G.Scabra and check the anti-tumor active of compounds by MTT.In this resrarch, the dried roots of the plant were extracted with hot methanol to give an extract, which was suspended in water and extracted with petroleum ether, ethyl acetate and n-butanol. The active sites of the three was separated by different chromatographic methods, such as Silicagel column, RP-18, Sephadex LH-20and so on. Eleven compounds were isolated from G. Scabra and identified by1H-NMR,13C-NMR and physicochemical methods. Their structures were identified as kaemp-ferol (1), erythrecentaurin (2), gentiaphyllide-D (3), myrianthic acid (4), gentiopicro side (5),6’-O-β-D-glucosyl-gentiopicroside (6), swertiamarin (7), loganic acid (8),6’-O-(2-hydroxy-3-O-β-D-glucopyranosyl-benzoyl)-sweroside (9). Compounds2,3,4,6and9are first isolated from G. scabra Bunge. The MTT experiments show that the Compound4and9have a toxic effect on Hep G2. Their IC50are199.37μM and149.85μM.
