Hepatitis B virus (HBV) infection causes acute and chronic hepatitis B,mainly through blood and blood products, the damaged skin and sexuallytransmitted, have close relationship with liver cirrhosis and primaryhepatocellular carcinoma, a great threat to human health, there is no specifictreatment. Looking for anti HBV monomer compounds from Chinese herbalmedicine is an effective method to found that HBV drug resistance.Jasminum officinale L.var. grandiflorum MuXi secco jasmine plant for J.Officinale dry flower bud, also known as mingyuan flower, jasmine, with liverXieYu effects. Widely cultivated all over the world, folk in the treatment of theduodenal bulb ulcer and chronic hepatitis, indigestion, and cirrhosis of theliver disease[1]. Zhao Guiqin[2]research shows that the main chemicalcomponents of Jasminum officinale L.var. grandiflorum iridoid glycosides andtriterpenoid saponins compounds.Iridoid glycosides has a variety of biologicalactivity, as a natural drug and the active components of traditional Chinesemedicine is commonly used as a cough medicine, fall blood pressure medicine,antipyretic and skin disease treatment.Hao ting[3]studies on the total iridoid glycoside extraction of Jasminumofficinale L.var. Grandiflorum,the results showed that ethanol extracted canmake the total iridoid glycosides purity surpass40%. In order to improvepurityof the total iridoid glycoside, the experiment adopts the macroporousadsorption resin purification technology for its enrichment, and screening thebest macroporous resin model, and optimizing its enrichment and purificationprocess. In the determining macroporous resin purification process, theisolated from the total iridoid glycoside of Jasminum officinale L.var.Grandiflorum and identification of structure, and detection anti-HBV activity of the monomer compounds, which provides the basis for further study, andanti HBV drug development to lay the foundation.The first partOptimization of enrichment and purification technology for total iridoidglycosides from Jasminum officinale L.var. gradiflorum by macroporous resinObjective:To study on enrichment and purification technology of total iridoidglycosides from Jasminum officinale L.var. gradiflorum with macroporousresin.Methods:With oleuropein as index, the content of it was determined by HPLC.Types of macroporous resin were optimized by static adsorption method,Enrichment and purification technology of it was investigated by orthogonaltest and single factors test.Results:XDA-16macroporous resin showed better effect than others, Theparameters were as follows: the concentration of liquid0.08g mL-1, height-diameter ratio of1:3, absorption velocity2BV h-1, dosage of sample liquidwas0.7g g-1, eluted by8BV water, the eluant volume of70%alcohol was4BV and elution velocity was2BV h-1.Conclusion:This optimized technology was stable and feasible. XDA-16macroporous resin had a good enrichment and purification effect on totaliridoid glycosides.The second partStudy on anti-HBV active constituent of Jasminum officinale L.var.grandiflorumObjective:To study on Study on anti-HBV active constituent of Jasminum officinale L.var. grandiflorumMethods:Isolation and purification were performed to silica gel column chromato-graphy and HPLC, the structures were elucidated on the basis of spectralanalysis and chemical methods. Using the means of HepG22.2.15cell modeland employing inhibition activity of HBeAg and HBsAg as the anti-HBVactivity-detecting marker.Result:Three iridoid glycosides were identified:6-O-methy-catalpol(1)、deacetyl asperulosidic acid(2)、loganin(3).6-O-methy-catalpol and deacetylasperulosidic acid inhibited the secretion of HBsAg, with IC50data86.8、67.9μg/ml.Conclusion:Compounds1~3were isolated from Jasminum officinale L. var.grandiflorum for the first time. The anti-HBV activities of Compounds1,2were discovered from Jasminum officinale L. var. grandiflorum for the firsttime.
