Objective:To observe the effect of Total saponin from Allium macrostem onBunge on animal models of Tail suspension test (TST),forced swimming test(FST),reserpine reversal test and with chronic stress depression. Determinate therelevant behavioral indicators of TST and FST,body temperature akinesia and eyelidptosis of reserpine reversal test.Meanwhile determinate the sugar consumption andweight,the activity of forced swimming test(FST),5-hydroxytryptamine (5-HT),dihydroxyphenyl ethylamine(DA) and norepinephrine (NE) content in brainhomogenate, levels of adrenocorticotropic hormone (ACTH) and corticosterone(CORT),related organ indexand and pathological morphous of tissues in mice modelwith chronic stress depression.Thereby investigate the therapeutic action andfeatures of Total saponin from Allium macrostem on Bunge on depression disease.Methods: Copy depression mice model by forced swimming test(FST) and tailsuspension test (TST).From the frist day of of test, grant Fluoxetine capsule(10mg·kg-1), high, middle and low dose of Allium macrostem on Bunge (300mg·kg-1,150mg·kg-1,75mg·kg-1), dissolve each of the corresponding drug in0.6%sodiumcarboxymethyl cellulose to intragastric administration,blank group give the samevolume of distilled water, once a day, continue7d. After1hours of the lastadministration,determinate the number of locomotor activity of mice in5minutes,the fixed time of mice within2~6min by the6min forced swimming test and tailsuspension test.Copy reserpine-antagonism test in mice.From the frist day of of test, grantFluoxetine capsule (10mg·kg-1), high, middle and low dose of Allium macrostem onBunge (300mg·kg-1,150mg·kg-1,75mg·kg-1), dissolve each of the corresponding drug in0.6%sodium carboxymethyl cellulose to intragastric administration,blank group and model group give the same volume of distilled water, once a day, continue7d.After1hours of the last administration,by peritoneal injection ofreserpine(4mg/kg),determinate the physical signs such as blepharoptosis,akinesiaand hypothermia.Rats were housed alone and exposed to an unpredicted sequence of mildstressors to induce CUMS to copy depression mice model.Divide blank group into2groups,5mice/cage, rearing model mice into single-cage, and accept irritationevery day, totally21days,from the frist day of test, grant Fluoxetine capsule(10mg·kg-1), high, middle and low dose of Allium macrostem on Bunge (300mg·kg-1,150mg·kg-1,75mg·kg-1), dissolve each of the corresponding drug in0.6%sodiumcarboxymethyl cellulose to intragastric administration,blank group give the samevolume of distilled water, once a day, continue21d.Determinate the sugarconsumption and weight,the fixed time of mice within2~6min by the6min forcedswimming test,the mice’s thymus, spleen wet weight and calculate organ index, take1/2brain preparation of brain tissue homogenate, determine5-HT, DA,NEcontent inbrain tissue,levels of adrenocorticotropic hormone (ACTH) and corticosterone(CORT),and observe hippocampus,hypothalamus,pituitary,thymus,spleen andadrenal gland morphological changes by microscope.Results:Depression model mice of TST and FST have been copiedsuccessfully.The fixed time of mice within2~6min by the6min forced swimmingtest and tail suspension test in model group was increased significantly,but the Totalsaponin from Allium macrostem on Bunge groups was decreased significantly.Depression model mice of reserpine-antagonism test have been copiedsuccessfully.The body temperature and ptosis of model group in mice induced byreserpine was decreased significantly,but the Total saponin from Allium macrostemon Bunge groups relieved the decrease significantly.Depression model mice induced by CUMS have been copiedsuccessfully.The sugar consumption and weight, the mice’s thymus, spleenin index,the5-hydroxytryptamine (5-HT), dihydroxyphenyl ethylamine(DA) andnorepinephrine(NE) levels in brain homogenate of rats in model group was reducedobviously or significantly,the fixed time of mice within2~6min by the6min forcedswimming test, adrenocorticotropic hormone (ACTH) and corticosterone (CORT)levels increased significantly. The Total saponin from Allium macrostem on Bungegroups could obviously or significantly improve the model rats’s sugar consumptionand weight, thymus, spleenin index,the5-hydroxytryptamine (5-HT),dihydroxyphenyl ethylamine(DA) and norepinephrine(NE) levels,decrease the activity of FST model rats,adrenocorticotropic hormone (ACTH) and corticosterone(CORT) levels.Conclusion:Each dose of Total saponin from Allium macrostem on Bungegroup could not only against the fixed time of mice of TST and FST,the bodytemperature and ptosis, also adjust the imbalance of5-HT,DA,NE,ACTH and CORTlevels, enhance immune function, enhance or restore the hymus,spleen,adrenalgland,hippocampus,hypothalamus,pituitaryt nerve cells in CUMS model mice.
