Objective:To approach the effects of Trichosanthes Kirilowii Maxim andAllium Macrostemon Bge (TKAM) on anti acute myocardial ischemia inrats, and to explore the role of MAPKs in acute myocardial ischemia.Method:Rats were divided into5groups randomly,set the TKAM group andTK,AM group,compared with Sham group and I/R group, ligationcoronary artery of the left anterior descending (LAD) cause acutemyocardial ischemia in rats damage model. Respectively tophysiological saline, Trichosanthes Kirilowii Maxim, AlliumMacrostemon Bge, and compatibility of TKAM to intervene14d, toobserve the change of S-T segment were observed; determination thelevels of maleic dialdehyde (MDA), superoxide dismutase (SOD), theactivity of the lactate dehydrogenase (LDH) and the activity ofcreatine kinase (CK) in serum, by tetrazolium chloride (TTC)Staining, the areas of myocardial infraction were observed; andthe cardiac muscle tissue was stained by HE to observe its necrosisarea and pathological changes.In addition, use the Western blotting method observationMitogen activated protein kinase (Mitogen activated protein kinases, MAPKs) signal path Extracellular signal conditioningkinase (Extracellular signal-regulated kinase, ERK), C-jun aminoend kinase (C-jun N-terminal kinase, JNK), p38MAPKsphosphorylation level of influence, validation MAPKs signal inacute myocardial ischemia induced myocardial damage occurs in thedevelopment of the role.Result:The TKAM group compared with other groups,(1) The TKAM groupcan be against acute myocardial ischemia after stage of theelectrocardiogram of S-T drive up, there are obvious difference andcompared with model group (P <0.01);(2) Compared with model group,I/R group of serum CK and LDH activity and MDA content increasedsignificantly (P <0.01or P <0.05), serum SOD activity decreasedsignificantly (P <0.05); TKAM group can obviously reduce serum CKand LDH activity and MDA content in rats, increased SOD activity(P <0.05or P <0.01);(3) Compared with model group, medicationgroup, namely: the TK group,AM group and TKAM group, all can reduceinfarction percentage significantly, and infarction percentagereduced, in turn and clear difference between each group (P <0.01);(4) By electron microscope and light microscope, according to theresults, Trichosanthes Kirilowii Maxim and Allium Macrostemon Bgecan improve the condition of myocardial cell fibers arrangeddisorder, reduce myocardial cell interstitial edema, reducebetween the muscle fiber inflammation cells infiltration,myocardial cell degeneration and necrosis;(5) TrichosanthesKirilowii Maxim and Allium Macrostemon Bge can effectively inhibitacute myocardial ischemia of rats myocardial cell damage, one ofthe mechanisms associated with the activation of MAPKs signaling pathways.Conclusion:Trichosanthes Kirilowii Maxim and Allium Macrostemon Bgeafter the compatibility, resistance to the effect of the myocardialinjury caused by acute myocardial ischemia is more pronounced, theeffect of the myocardial injury caused by electrocardiogram (ecg)against acute myocardial ischemia model of rat S-T segmentelevation have more apparent confrontation effect, can reduce theactivity of LDH and CK in serum and MDA content, increase serum SODactivity, the obvious myocardial infarction narrow scope, protectmyocardial tissue; Three kinds of wire break original activatedprotein kinases (MAPKs) in Trichosanthes Kirilowii Maxim and AlliumMacrostemon Bge play different role in the fight against acutemyocardial ischemia.
