Studies On Chinese Medicine Allium Macrostemon Bunge As An Effective Treatment For Chronic Myocardial Ischemia

Myocardial ischemia(MI)is a kind of cardiovascular diseases,which go along with the damage of heart cells and the decrease of oxygen and blood supply,and it becomes the significant enemy on human health.Chronic myocardial ischemia(CMI)is a kind of MI,which presents high incidence in recent years.Traditional Chinese medicine(TCM),Allium macrostemon Bunge(AMB),has been used in treating CMI clinically and demonstrates good efficiency,while its therapy mechanism is still unknown.Metabonomic is a systematic method focusing on the profile of low molecular weight metabolites in biofluids and tissues and their response to disease and toxicological.In this thesis,a combination of nuclear magnetic resonace(NMR)spectroscopy and multivariate statistical analysis method was used to analyze the metabonomic changes of mouse in normal,isoproterenol-induced CMI and AMB-treated groups.The aim is to find the detailed metabolic response of CMI and the therapeutic effect of AMB.The specific content is shown as follow:First,1H NMR spectra of rat urine have been recorded for the normal,CMI and AMB-treated group from different time.Based on the combination of multivariate statistical analysis and biochemical data,the results show(1)CMI aroused metabolic disorders,such as energy metabolism,effect of gut microbiota,Ca2+ overload,and so on.However,AMB could recover the disorders at a certain extent.(2)The therapeutic effect of AMB might rely on the time that the CMI patients are treated with AMB,its efficacy brought out at maximum extent with continuous infusion of AMB,and this was confirmed to the TCM theory.Second,from the electrocardiograms and histopathological assessments,together with NMR-based metabonomic profiles of rat heart,liver and kidney in normal,CMI and AMB-treated group.The results present as follow:(1)The volt of ST wave in MI rats is higher which is a primary factor on MI and myocardial infarction diagnosis.(2)The cells of heart have been damaged severely,and a large number of inflammatory cell infiltration appears.(3)CMI caused several metabolic disorders,such as energy metabolism,amino acids metabolism,choline metabolism,nucleotide metabolism,and so on.However AMB has an obvious repairment to the CMI-induced series of disorders.
