Deer blood is chamber blood or chopped blood of Cervus nippon or Cervus elaphus, which is our traditional tonics and has several pharmacological activities include anti-aging,immune stimulation, antifatigue and other functions. Since blood chopped is less, so generallythink that deer blood is chamber blood. But now the relationship between chemicalcomposition in deer blood and its function is not yet clear, therefore, in this paper, differentsolvents were used to extract, a preliminary study was done to study deer blood relatedfunctions, then bioactive extracts were isolated and clarified by diversified columnchromatography, seven compounds of deer blood were got, incorporated with NMR andrelated literature, then we identified the structures of the compounds. With alcohol, petroleum ether and other solvents, by some steps to get the correspondingfour deer blood extracts. D-galactose was used to make aging model. Through comparingMAO activity in brain tissue in mice, MAD content which in liver tissue of control group andexperimental group, we can see that: in comparison with model set, deer blood petroleumether extract, alcohol extract group, MAO activity which in brain dropped remarkably (P <0.05), MDA content which in liver dropped highly significant (P <0.01); But in comparisonwith model set, deer blood water and aqueous extract-alcohol sedimentation group, MAOvitality in brain with MDA content in liver tissue had no significant changes (P>0.05). Sincewe can conclude, deer blood petroleum ether extract and alcohol extract both can make MDAcontent which in mice liver tissue and MAO activity which in brain all decreased significantly, with the effects of oxidation resistance and age defying.By comparing the deer blood extract on the influence of the experimental group andcontrol group mice swimming time, the results showed that the deer blood petroleum etherextract group mice swimming exhaustion significantly prolonged (P <0.05), in comparisonwith control group, the anti-fatigue ability was even taller. Research findings showed that thedeer blood petroleum ether extract group exercised fatigue resistance of the best effect.Immune improving of experiment by comparing the deer blood extract IgG content andratio of viscera weight and body weight of the experimental group and model group mice. Incomparison with model set, serum IgG content in mice in the deer blood alcohol extractgroup had significantly increased (P <0.01), but in comparison with model set, serum IgGcontent in mice in deer blood water extract, light petroleum extract and aqueousextract-alcohol sedimentation group had no marked discrepancy(P>0.05), showed that deerblood alcohol extract group had improved serum IgG content in mice; In comparison withmodel set, spleen and thymus index in mice had extremely significant difference in deerblood alcohol extract(P<0.01), showed that deer blood alcohol extract have the role ofincreasing spleen and thymus index in mice. Since we can conclude that deer blood alcoholextract has a role in the increase of spleen and thymus index, serum IgG content in mice, canraise the body’s immune ability to a certain extent.we knowed that deer blood alcohol extractive and petroleum ether extract withoutstanding anti-aging, anti-fatigue and immune improving activities, so they were got into MCI column on merger, divided into A, B, C three parts, then each separated by discrepantchromatographic columns, finally, the compounds in A part is:(1)α-ethoxy-mannose; threemain compounds in Part B are:(2)uracil;(3)adenosine;(4)inosine; three main compounds inPart C are:(5)4,14-male ene-2alcohol-28,30cyclopentadiene heptanoic acid ester-3ketone;(6)4,14-male ene-2alcohol-27,29cyclopentadiene has ether-3ketone;(7)4,14-male ene-2,12alcohol-28,30cyclopentadiene heptanoic acid ester–3ketone.
