Fatigue is a comprehensive reaction of various physiological and biochemical changes in the body after a long period of exercise,which can affect people's quality of life to varying degrees.If fatigue is not restored in time,it will lead to "overwork",overtraining syndrome,chronic fatigue syndrome,etc,causing endocrine disorders,decreased immunity,and even organic lesions.An important factor threatening human health.Currently commonly used treatment methods include behavioral cognitive therapy,antioxidants,and herbal extracts.Injection of Deproteinized Culf Blood Extractives Injection(DCBEI)is a sterile biological preparation rich in amino acids and trace elements.It has strong pharmacological action and is often used to treat neurological deficits,skin burns,gastrointestinal diseases,arterial vascular diseases,etc.It has been confirmed to have an antioxidant effect,and its research on acute and chronic fatigue has not been reported.Therefore,we will explore the therapeutic effects of DCBEI on acute and chronic fatigue and explore relevant mechanisms to provide further data support and ideas for the development of acute and chronic fatigue.The research contents are as follows: 1.Analysis of main components of DCBEIThe component having a molecular weight of 500 or less as determined by gel permeation chromatography(GPC)accounts for 93.92%,and the molecular weight is small,which is favorable for absorption by the body.The Deproteinized extract of calf blood for injection,which is determined by high performance liquid chromatography(HPLC),is rich in amino acids and nucleotides,which are the main raw materials for the synthesis of proteins and genetic material(DNA and RNA).It can provide abundant raw materials for body tissue construction and cell regeneration.2.Study on the anti-acute fatigue effect of DCBEIHealthy BALB/c male mice were administered with DCBEI for 18 days,and subjected to autonomous activity test,running test,weight-bearing forced swimming test and rotary rod test,and then tested in mouse serum and liver.Biochemical indicators of fatigue and oxidative stress.Animal behavioral experiments showed that DCBEI can improve the running time,fatigue turning time and swimming time of acute fatigue mice,but had no significant effect on the spontaneous activity and standing of mice.By detecting the biochemical indicators of acute fatigue mice,DCBEI can increase the ATP content in serum,muscle and liver of acute fatigue mice,and significantly inhibit the expression level of LDH.DCBEI also increases the expression levels of muscle glycogen in the muscle and liver glycogen in the liver.In addition,DCBEI is able to modulate oxidative stress in serum,muscle,and liver of acutely fatigued mice,such as ROS and MDA in mice serum,muscle,and liver.These indicate that DCBEI has a certain regulatory effect on acute fatigue mice and enhances its anti-fatigue activity,which may be related to oxidative stress.3.Study on the anti-chronic fatigue effect of DCBEIThe BALB/c mice were subjected to cold water swimming,fatigue running,fatigue rotating rod and sleep deprivation.The animal behavior test stimulated the establishment of a chronic fatigue mouse model,and then the DCBEI was administered by intragastric administration to investigate the anti-chronic fatigue effect of the mice.And preliminary mechanism research.After DCBEI administration,it can significantly improve the autonomic activity,running time,fatigue turning time and weight-bearing forced swimming time of chronic fatigue mice.In addition,the detection of relevant biochemical indicators in chronic fatigue mice showed that DCBEI can promote the expression of ATP in muscle and liver of chronic fatigue mice,and can reduce the expression levels of LDH and BUN.DCBEI can also increase the expression levels of muscle glycogen in the muscles of chronically fatigued mice and liver glycogen in the liver.Finally,DCBEI also significantly inhibited the levels of oxidative stress,such as ROS and MDA,in serum,muscle and liver of chronically fatigued mice.These preliminary indications that DCBEI can increase the endurance of chronically fatigued mice and increase the anti-fatigue activity of mice.In summary,DCBEI has certain anti-fatigue activity in acute fatigue mice and chronic fatigue mice,and improves the endurance of mice.The mechanism of action may be related to DCBEI's ability to modulate oxidative stress in mice.The results of this study suggest that DCBEI may be a good candidate for the development of new anti-fatigue functional supplements.
