Objective: To investigate the expression of transforming growth factor-β2 (TGF-β2) in human vitreous from patients with proliferative diabetic retinopathy ( PDR ) or proliferative vitreoretinopathy ( PVR).To study the possible effects and mechanisms of TGF-β2 on proliferative vitreoretinopathy patients .Methods: 15 vitreous specimens were collected from the eyes of patients with PDR undergoing virectomy,35 vitreous specimens from the eyes of patients with PVR ,and 17 vitreous specimens from the normal donated eyes served as controls. Protein expression of TGF-β2 from the vitreous of the eyes with PDR and PVR were assessed by Western blot analysis. Both total and mature protein expression TGF-β2 were quantified.Results:①The total protein expression of TGF-β2 showed significant difference between PDR group(20.43±6.37)and the control group(7.57±1.35,P<0.01);And significant difference between PVR group(13.43±2.97) and the control group.②The mature protein expression of TGF-β2 showed significant difference (P<0.01)between PDR group (4.06±1.43)and the control group(0.56±0.28);And significant difference between PVR group(2.19±1.31) and the control group .③There were no statistic differences of the mature TGF-β2 between PVR B stage and the control (P﹥0.05),but significant difference between the control group and other subgroups ( P<0.05 ) .④No significant correlationship were found between TGF-β2 ,and age ,sex etc factors. There was correlation between TGF-β2 and severity of the disease.Conclusion: There are abnormal protein expressions of TGF-β2 protein in vitreous derived from the eyes with PDR and PVR patient, TGF-β2 may play an important role in developing PDR and PVR. Furthermore, the protein expression level of TGF-β2 changes in different PDR and PVR stages, which suggest the different effects and mechanisms in the proceeding of PDR and PVR.
