Objective: Investigate the effect of Corydalis saxicola Bunting onproliferation and apoptosis and its possible mechanism of human lung cancerA549cell.Methods: The proliferation of lung adenocarcinoma cell line A549cultured in vitro were divided into4groups randomly:1The control group,2The Corydalis saxicola Bunting group,3The Cisplatin group,4The Corydalissaxicola Bunting and Cisplatin Combined treatment group. MTT method wasused to investigate the proliferation of Corydalis saxicola Bunting, cisplatin andcombined with each other. MTT assay was used to detect the proliferation ofCorydalis saxicola Bunting in different concentrations (0.1,0.05,0.01,0.005g/L)respectively effect24h,48h,72h.Hoechst stain was applied to analyze theapoptosis effects. Changes of caspase-3and survivin mRNA expression aftertreatments were detected by RT-PCR.As the changes of protein expression weredetected by Western-blot.Results: Corydalis saxicola Bunting could inhibit the proliferation of A549 cells in dose-time relationship. After48h, inhibition of combined group in A549cells is stronger than single drug group (P<0.05). Morphological observationshowed that the number of apoptotic cells in the combination group more thanthe single drug group. RT-PCR results suggest that compare with the single druggroup,combination group raised the caspase-3mRNA and down-regulated theexpression of survivin mRNA(P<0.05).The trend of protein expression are samewith the mRNA expression.Conclusions: Corydalis saxicola Bunting can inhibit the proliferation ofA549cells. Combining with cisplatin can enhance its inhibitory effect on A549cells, and inducing apoptosis in cancer cells. The mechanism may be related tothe up-regulation of caspase-3expression and down-regulation of survivinexpression. This study explored the clinical treatment of lung cancer treatmentideas and a new combination of drugs, laid the foundation for further study.
