Objective:to study the expression of ROS tyrosine kinase in cholangiocarcinoma and cholangiocarcinoma cell lines, and the relationship between the expression of ROS and clinical indicators. To provide a theoretical basis for further exploring a new molecular target for cholangiocarcinoma diagnosis, treatment or prognosis.Method:Immunohistochemistry was used to detect the expression of ROS in the cancer tissue and adjacent tissue of50cases confirmed by the clinical pathological experts of the Third Xiangya Hospital, Central South University, and the relationship between the expression of ROS and clinical indicators (the patient’s gender, age, location, differentiation degree, lymph node metastasis, TNM stage) relationship. Western blot and RT-PCR was used to detect the expression of ROS in HuCC-T1cells, RBE cells and cell QBC939, the three lines of cholangiocarcinoma cells, and fresh cholangiocarcinoma.Results:ROS highly (the positive expression rate is50%) expressed in cancer tissues in cholangiocarcinoma but lowly (the positive expression rate is3%) expressed in adjacent cancer tissues, P<0.05; the highly expression of ROS was correlated with lymph node metastasis (the positive expression rate of lymph node metastasis was61.8%, and no positive lymph node metastasis was25%, P=0.0246<0.05) and TNM stage (the positive expression rate of stage I and II was25.0%, and the positive expression rate of stage III was67.7%, P=0.03570.05), but not correlated with the patient’s gender, age, location and differentiation degree; the expression of ROS in Hu-CCTl cells was positive, but in RBE cells and cell QBC939was negative.Conclusion:ROS tyrosine kinase may be involved in the occurrence of cholangiocarcinoma. ROS tyrosine kinase may be involved in the invasion and metastasis of cholangiocarcinoma. The Hu-CCT1cell line may be used as a represent to do the next biology experiments to future investigate the role of ROS tyrosine kinase in cholangiocarcinoma.
