| ObjectivesTo explore the molecular mechanisms underlying traditional Chinese medicine (TCM)for the treatment of patients with obesity and provide pharmacological evidence forclinical practice, we identify the TCM with the ability to inhibit the expression ofplasminogen activator inhibitor-1(PAI-1) gene.MethodsThe experimental protocols are described below. Firstly, cell morphology and MTTassay were used to evaluate the changes in the proliferation of mouse3T3-L1cells treatedwith the extracts of six kinds of Chinese proprietary at the concentration of0.01,0.1,1, or10mg/ml for24hours. Based on the results, we determined the appropriate concentrationat which the extracts of the TCM have minimally effects on cell morphology with thegreatest effect on cell proliferation. Secondly, quantitative real-time PCR was used todetermine the effects of the extracts of various Chinese proprietary on the expression ofPAI-1gene in3T3-L1cells.Results1Rests of MTTBased on the results of morphology and MTT assay, we found that Differentconcentrations may be inhibited to varying degrees ECMH of3T3-L1precursorproliferation, The1mg/ml is the highest concentration inhibition rate except XiaoChengqiserum. Compared with normal control group, All concentrations of FangfengTongshengSanã€Fangji Huangqi serumã€Jianfei Tiaozhi Jiaonangã€ZeXie serum are havestatistically significant(P≤0.05),beside the10mg/ml of ShenLing BaiZhuSan,the 100ug/mlã€1ug/ml of XiaoChengQi serum, the other Concentration are all have statisticallysignificant(P≤0.05).2Rests of MTTUsing quantitative Real-time PCR, we found that the expression of three differentconcentrations on six Chinese herbs on PAI-1gene in3T3-L1cells are all decreased.Compared with the control group,the expression of PAI-1gene ofShengLingBaiZhuSanã€Fangfeng TongshengSanã€XiaoChengqi serum are all havestatistically significant, beside the10mg/ml of Jianfei Tiaozhi Jiaonangã€FangJi HuangQiserum,1mg/ml of Zexie serum, the other Concentration are all have statisticallysignificant(P≤0.05).Conclusions1.Different concentrations may be inhibited to varying degrees ECMH3T3-L1precursorproliferation, Extent of inhibition correlated with the concentration of the formulation.2.Different concentrations may be inhibited the expression of3T3-L1cells of PAI-1,andthus may affect the obesity gene expression, inhibit the secretion of3T3-L1cells and atlast achieve weight loss goals. |
