Effect Of Study On The Yiqi Capsule On BPH Miceand Rats Models

Objective:To preliminarily reveal that Yiqi capsule can cure hyperplasia(BPH) by measuring the prostate weight and the prostate exponent of mice and rats models, and observing the serum and the level of testosterone(T) and dihydrotestosterone(DHT) and estradiol(E2) and prostatic acid phosphatase(PACP), morphological changes of the prostate, and investigating the efficacy of Yiqi capsule on BPH.Methods:To establish BPH model of rats by subcutaneous injecting testosterone propionate induction method. With successful modeling rats models were divided into seven groups:control group, model group, Longbishu group (0.3g/kg Longbishu suspension), finasteride group (0.833mg/kg finasteride suspension), Yiqi capsule low dose group (0.03375g/kg Yiqi capsule suspension), middle dose group (0.0675g/kg Yiqi capsule suspension) and high dose group (0.135g/kg Yiqi capsule suspension), and control group and model group were given the same amount of solvent,1times a day, then treated with each group’s drug at the same time for30days. Rats were castrated, from the eighth day subcutaneous injected testosterone propionate4mg/kg (soluble in soybean oil) for30days. After two hours, rats were given drugs last time, pick out the eyeball to get blood, then were observed the prostate weight and the prostate exponent, and measured the level of DHT andE2and PACP, and observed morphological changes of the prostate.The mice model of BPH was also established by urogenital sinus implantation. With successful modeling mice models were divided into seven groups:control group, model group, finasteride group(1.266mg/kg finasteride suspension), Longbishu group(0.15g/kg Longbishu suspension), Yiqi capsule low dose group(0.1g/kg Yiqi capsule suspension), middle dose group(0.0675g/kg Yiqi capsule suspension) and high dose group(0.2g/kg Yiqi capsule suspension), and control group and model group were given the same amount of solvent,1times a day, then treated with each group’s drug at the same time for21days. After2hours, mice were given drugs last time, pick out the eyeball to get blood, then were observed the changes of the prostate weight and the prostate exponent, and observed the level of PACP and DHT, and observed morphological changes of the prostate, the thymus and the spleen.Results:The BPH mice and rats models were all successful modeling. Yiqi capsule can significantly decrease prostate weight and prostate exponent; high dose and low dose of Yiqi capsule can significantly decrease prostate weight and prostate exponent; high dose, middle dose and low dose can significantly decrease the content of DHT and T and significantly increase the content of E2; high dose and low dose of Yiqi capsule can significantly decrease the level of PACP. High dose, middle dose and low dose of Yiqi capsule can significantly decrease the prostate weight and the prostate exponent of mice model, and significantly decrease the level of DHT(P<0.01) and the level of PACP.Conclusion:Yiqi capsule can significantly decrease the prostate exponent, the prostate weight, the content of DTT and T, and significantly increase the level of E2of mice and rats model. It is shown that Yiqi capsule can cure BPH mice and rats.
