Objectives1: To establish rat model of prostate hyperplasia,to screen the best time point for the model production of Castrated rats' Prostatic Hyperplasia Induced by Testosterone Propionate.2: To investigate the effect on rat prostate and its possible mechanism when the models above are injected with murine HMGB1 protein.Methods1: Forty-eight adult male SD rats were randomly divided into 6 groups,eight rats in each group.The groups shall be named as Control group,Castration control group,7d group(7 days after subcutaneous injection of testosterone propionate),14 d group,21 d group,28 d group;Measure the body weight,prostate volume,wet weight and prostate index;Observe the histopathological changes of prostate tissue,through the use of HE staining;Detect the expression of HMGB1,RAGE and TLR4 in rat prostate tissue by using immunohistochemistry and Western blot.2: 32 adult male SD rats were randomly divided into 4 groups,8 in each group: named as 21 d model group,Low dose group(castrated to the rat prostate tissue after injection with 100 mg murine HMGB1 protein,while 21 days after subcutaneous injection of testosterone propionate),Medium dose group(200 mg),High dose group(400mg);Measure the body weight,prostate volume,wet weight and prostate index;Observe the histopathological changes of prostate tissue,through the use of HE staining;Detect the expression of HMGB1,RAGE and TLR4 in rat prostate tissue by using immunohistochemistry and Western blot.Results:1: Atrophy of rats' prostate tissue emerges after castration,Compared with those of the control group and 7d group,the prostate volume,prostate wet weight and prostate index of the rats in the 14 d,21d and 28 d groups were significantly increased(P <0.05).Moreover,the volume of prostate in group 21 d and group 28 d was significantly higher than that in 14 d group(P <0.05).But there was no significant difference between those in the 21 d group and the 28 d.Rat prostate epithelial cells and interstitial tissue changes in proliferation in 14 d group;Rat prostate epithelial cells and interstitial tissue showed significant proliferation,and inflammatory cell infiltration phenomenon emerges in 21 d group and 28 d group;Compared with the control group,the expression of HMGB1,RAGE and TLR4 in the prostate tissue of the 21 d group increased(P <0.05).Compared with the control group,the expression of HMGB1 and TLR4 in the prostate tissue of the 28 d group increased(P <0.05).Compared with the 21 d group,HMGB1 middle dose group and high dose group index increased significantly(P <0.05),no significant difference between the other groups;Compared with the 21 d group,the expression of HMGB1,RAGE and TLR4 in the HMGB1 high dose group was significantly increased(P <0.05),but there was no significant difference between the other groups.Conclusion:1: The method of subcutaneous injection of testosterone propionate after castration,Rat model of benign prostatic hyperplasia can be successfully constructed in 21 days,and in the process accompanied by HMGB1,RAGE,TLR4 expression increased.2: The method of injecting medium and high dose recombinant mouse HMGB1 protein in ovariectomized model of ovariectomized rats,the rat prostate index can significantly increase,and in this process accompanied by HMGB1,RAGE,TLR4 expression increased.
