Background and ObjecktiveCondyloma acuminatum(CA) is one of the common sexually transmitteddisease(STD) at present. In our country, the incidence of CA occupies the secondplace of STD, and showed a trend of increasing year and year. The disease which is abenign proliferative is caused by human papillomavirus (HPV) infection, that mainlygrows in the skin and mucous membrane such as the genital and anal. The majorclinical manifestation of the CA is cauliflower, papilloma, but it has proliferatedrapidly, infections strong, high recurrence rate and other characteristics, and somehuge CA has the possibility of malignant transformation, because of thesecharacteristics, CA brings great negative influence to the patient’s health and social,family, so its prevention and treatment has been concerned.So far the CA incidence of the exact pathogenesis is still not completely clear, inrecent years, mainly for the cellular proliferation and apoptosis, angiogenesis andcellular immunity and other aspects of its pathological mechanism is discussed, andhave achieved some research results. Wnt signaling pathway is recognized as animportant signal in the process of embryonic development system, especially thestudy on the Wnt/-catenin classic signal transduction pathway is more and moremature, the Wnt1protein as the ligand and starting protein is most common inclassical pathway, it is activated by combining corresponding receptor on the cell membrane, and inturn activation sequence transduction molecule, ultimately totranscription and translation of the downstream target genes (CyclinD1, C-myc) toadjust and control, with the role of regulating cell growth, cell development anddifferentiation. So the path a chain defects or abnormal expression of all can lead tobiological growth defects. In recent year found that this is closely related with theoccurrence and development of wide variety of tumor. Tumor suppressor in lungcancer1(TSLC1) is one of the newly discovered tumor suppressor gene, in the studyof different cancer tissues has confirmed its lower expression or absence, and now itmaybe related to cell adhesion and signaling functions, also closely related to tumorimmunity, cell apoptosis induction and the inhibition of cell growth cycle. In recentyears, the study of the two mainly concentrated in the respiratory tract cancer,digestive tract tumor, and little research in the pathogenesis of CA. This experimentwe use SABC immunohistochemical method in45cases of CA and24cases ofnormal prepuce tissue to detect the expression of Wnt1and TSLC1, and analyze thecorrelation and its significance between Wnt1and TSLC1in CA tissues, to furtherunderstand the pathogenesis of CA.Materials and Methods1Materials45cases of CA specimens as experimental group are collected from theoutpatient department of dermatology in our hospital, including26males,19females;Age18to60years old, average33.29years; Course of15days to120days, with anaverage of53.89days. All the cases with typical clinical manifestation are confirmedby histopathological diagnosis, and has not been accepted any treatment before thedrawn. The control group consisted of24patients with department of urologyoperation resection of adult male of normal skin tissues, and confirmed byhistopathology.2Methods45cases of CA group and24cases of normal prepuce, immunohistochemicalSABC method were applied to detect Wnt1, TSLCl in expression of these cases, thespecific steps in strict accordance with the kit instructions. Wnt1positive stainingmainly locate in the cytoplasm, some can be found in the cell membrane, visible light yellow to brown granule;TSLC1positive color locate in the cytoplasm and cellmembrane,which together with pale yellow to brown granule.3Statistical processingUsing SPSS17.0statistical software to analyze the data, in the experimentalgroup and control group the positive expression rate difference of Wnt1and TSLClapplication2test are analyzed; Expression intensity differences between the twogroups applied rank sum test. The CA group correlation between two variables wereanalyzed by the Spearman correlation of non parametric system, P <0.05for thedifference was statistically significant.Results1The Wnt1expression in CA and normal prepuce tissueIn45cases of CA tissues the positive expression of Wnt1is37cases, the rate ofpositive expression is82.22%(37/45).The intensity of expression is+++++, thepositive staining of Wnt1is mainly located in the cytoplasm; The positive expressionof Wnt1in24cases of normal prepuce tissue is7cases, the rate of positiveexpression is29.17%(7/24).The intensity of expression is-+. The difference ofpositive expression rate between the two groups has statistical significance(2=19.070, P<0.05), and positive expression intensity differences between the twogroups are statistically significant (H=21.131, P <0.05).2The TSLC1expression in CA and normal foreskin tissueIn45cases of CA tissues the positive expression of TSLC1is17cases, thepositive expression rate is37.78%(17/45).The intensity of expression is+++, thepositive staining of Wnt1is mainly located in the cytoplasm; The positive expressionof TSLC1in24cases of normal prepuce tissue is22cases, the rate of positiveexpression is91.67%(22/24), expression intensity is+++. The positive expressionrate of the two groups has significant statistical significance(2=18.496,P<0.05), andthe differences of positive expression intensity between the two groups arestatistically significant (H=34.770, P <0.05). 3Correlation analysis of Wnt1and TSLC1expression in the CA tissuesIn45cases of CA tissues, the positive expression of Wnt1is37Cases andnegative is8Cases;in TSLC1expression,17cases are positive,28cases arenegative.Pearson correlation analysis found that45cases of Wnt1and TSLC1expression was negatively correlated(r=0.336,P <0.05).Conclusions1. The positive expression of Wnt1in CA tissues is higher than the control group,it may affect the apoptosis and proliferation of cells and plays an important role to thegrowth process of CA.2. The positive expression of TSLC1in CA tissues is lower than the controlgroup, this kind of low abnormal expression may be by activating signaling pathwayinhibits cell apoptosis and immune escape, prompting CA abnormal growths of theorganization.3. Wnt1and TSLC1expression in CA group showed a negative correlation, thetwo may promote the occurrence and development of CA.
