Objective:Investigate the expression of miRNA-126and VEGF between the chorionic villus of early embryo growth arrest group and the chorionic villus of control group. Analysis the different expressions of miRNA-126and VEGF between them. Explore the relationship of miRNA-126and VEGF in early embryo growth arrest.Methods:1. With28cases of chorionic villus in early embryo growth arrest group and28cases of chorionic villus in control group, using the method of RT-PCR, the changes expression level of miRNA-126in the two groups were detected.2. With28cases of chorionic villus in early embryo growth arrest group and28cases of chorionic villus in control group, using the method of RT-PCR, the changes expression level of VEGF mRNA in the two groups were detected.3. With10cases of chorionic villus in early embryo growth arrest group and10cases of chorionic villus in control group, using the method of S-P immunohistochemical staining, the changes expression levels of VEGF protein in the two groups were detected.Results:LmiRNA-126expressed in the chorionic villus of early embryo growth arrest group and control group,which relative miRNA-126expression were1.1727±0.1824and1.0135±0.1750. The expression of miRNA-126in the chorionic villus of early embryo growth arrest group was higher than the control group, the difference was no significant (P>0.05).2. VEGF expressed in the chorionic villus of early embryo growth arrest group and control group, which relative VEGF expression were0.3350±0.0756and1.0299±0.1986. The expression of VEGF in the chorionic villus of early embryo growth arrest group was significantly lower than the control group, the difference was statistically significant (P<0.001).3.Through the SP immunohistochemical staining found that VEGF expressed in the chorionic villus of early embryo growth arrest group and control group. VEGF was mainly expressed in trophoblast cells (including cytotrophoblast and syncytiotrophoblast) and vascular endothelial cell cytoplasm. The average optical density of VEGF in the chorionic villus of early embryo growth arrest group (0.2847±0.0180)was significantly lower than the control group(0.4877±0.0254), the difference was statistically significant (P<0.01).4. Negative correlation was found between miRNA-126and VEGF in the chorionic villus of early embryo growth arrest group(r=-0.667,P<0.01). Positive correlation was found between miRNA-126and VEGF in the chorionic villus of control group(r=0.930,P<0.001).Conclusion:1. The expression of miRNA-126in the chorionic villus of early embryo growth arrest group and control group was no significant difference.2. The expression of VEGF in the chorionic villus of early embryo growth arrest group was lower than in the chorionic villus of control group.3. There are some differences in the correlation miRNA-126and VEGF in the chorionic villus of early embryo growth arrest group and control group,prompts the occurrence which VEGF the expression change and the early embryo growth arrest to have certain relevance.
