Objective:Early embryo growth arrest,with its high incidence and complicated etiology,is a disease that seriously affects women's physical and mental health.By collecting clinical case data and detecting the expression of long non-coding RNA HOTAIR and EZH2 in fetal villi during early gestation,this study explored the role of HOTAIR and EZH2 in the mechanism of embryo germination during early gestation,Providing a new experimental theoretical basis for the study on the pathogenesis of embryo germination during early gestation.Methods:This study collected the villi obtained during curettage under aseptic conditions from the abortion patients in the family planning outpatient department of the first people's hospital of zhangjiagang from October 2017 to February 2018 as experimental samples.The experimental group included 23 patients with confirmed embryo sterilization by B-mode ultrasound,and the control group included 24 patients with spontaneous termination of pregnancy at the same gestational age.All the enrolled patients had no systemic diseases in the past,denied the contact history of toxic substances,and the gynecological examination excluded genital tract inflammation.Both husband and wife had no chromosomal abnormalities.1.Statistical analysis was conducted on the general data and clinical test data of the experimental group and the control group:age,number of pregnancies,gestational sac diameter,blood coagulation function,reproductive endocrine hormone,thyroid function related hormone,etc.2.Total RNA extraction and purification were performed on the collected villus samples.Real-time polymerase chain reaction(RT-PCR)was used to detect the expressions of HOTAIR and EZH2mRNA in the villi of the experimental group and the control group.3.Immunohistochemistry was used to detect the localization and expression of EZH2 protein in villi of the experimental group and the control group.Results:1.There were no statistically significant differences in age,number of pregnancies,gestational sac diameter and blood coagulation function between the 23 patients in the early gestation embryo abortion group and the 24 patients in the early gestation normal embryo group(P>0.05).The results of reproductive endocrine hormone test showed that there was no statistically significant difference in FSH between the experimental group and the control group(P>0.05),but E2,P,and LH in the experimental group were significantly lower than those in the control group(P<0.05).Thyroid function related hormone test results showed that there was no statistically significant difference in FT3 and FT4 between the experimental group and the control group(P>0.05),while TSH in the experimental group was significantly higher than that in the control group(P<0.05).2.RT-PCR results showed that the expression of HOTAIR in villi of the experimental group was higher than that of the control group,but P>0.05.The expression level of EZH2 mRNA was significantly lower than that of the control group(P<0.05).Pearson correlation analysis showed that the correlation coeffieient between HOTAIR and EZH2 mRNA expression levels in embryo sterile villi was not statistically significant(r=-0.116,P=0.597>0.05).3.Immunohistochemical detection results showed that EZH2 protein in villi tissues was mainly stained by the nucleus,with some cytoplasmic staining.The expression intensity of EZH2 protein in villi tissues of the experimental group was significantly lower than that of the control group(P<0.05).Conclusion:1.Through the analysis of general data and clinical test data,this experiment verified that reproductive endocrine hormone and thyrotropin are closely related to embryo abortion.2.Expressions of HOTAIR and EZH2 mRNA can be detected in villi of embryo aborting and normal abortion.The expression of HOTAIR in villi of embryo aborting patients showed an increasing trend,while the expression of EZH2 was significantly decreased,which may be involved in the pathogenesis of early embryo aborting.3.There was no significant correlation between HOTAIR and EZH2 expression in the fetal villi,suggesting that HOTAIR may be involved in the occurrence of embryo shutdown by binding EZH2 to make functional changes in the polycomb repressiv complex2(PRC2)-dependent regulation pathway.
