Effect Of "Yangnao Prescription" On Neurological Impairment For The Rat Model With Permanent Focal Cerebral Ischemia In Early Recovery Stage

Objective:To observe the "Yangnao Prescription " effecting on behavior improvement for the rat model with permanent cerebral ischemia in early recovery stage, and discussing the neural functional recovery mechanism; providing a new Chinese medicine treatment for cerebral infarction patients in early clinical therapy.Methods:At first, the rat models with permanent middle cerebral artery occlusion(pMCAO) should be established. After24hours, the grade of the left paralyzed limbs were evaluated by Longa grading for rat models. All experimental animals were divited into sham operation group、blank goup、western medicine group and traditional Chinese medicine group by using a complete randomized grouping method in this study. Then, each group was given gavage once a day after3days-14days of operation. At the3th day、7th day、10th day and14th day, neurological deficits and bar walking test were evaluated again in every group. At the14th day, the experimental animals were executed after dose. The morphological structure in ischemic cerebral cortex were observed by hematoxylin eosin staining. The transcriptional level of HIF-la and VEGF was detected in the right ischemic cerebral cortex by employing RT-PCR. The expression of HIF-1α and VEGF were examined in the right ischemic cerebral cortex by employing the immunohistochemistry assay. All the data was analyzed by using completely random design multiple analysis of variance for statistical test.Result:1、Compared with model group,"Yangnao Prescription can obviously improve left posture response, and promot the coordinating ability of movement (P<0.01) for rats with cerebral ischemia;2、The expression of HIF-1β and VEGF in the right ischemic cerebral cortex in traditional Chinese medicine group and western medicine group were extremely higher than other groups (P=0.000, P<0.01); Compared with western medicine group, the expression of VEGF in traditional Chinese medicine group were higher (P=0.009, P<0.01)3、There was no statistical differences between traditional Chinese medicine group and western medicine group on transcriptional level of HIF-1α and VEGF (P>0.05).Conclusion:"Yangnao Prescription " can extremely improve neurological fuction recovery for the rat model with permanent focal cerebral ischemia in early recovery stage. The potential mechaism was related to activate HIF-1α, then promote the expession of VEGF.
