Objective: Dtective effect morphology of rat bone marrow DC, CD86、OX62on thesurface with different concentrations of sinomenine.Method:1, The cultivation of rat bone marrow DC precursor cells and imDCpurification.2, Group:(1) control group(2) the sinomenine group(3) LPS stimulation group(4) sinomenine+LPS stimulation group.3, With a microscope observation of DC morphology under differenttreatment groups4, Flow cytometry instrument testing CD86、OX62expression before andafter the training.Results:(1) It shows a small amount of increased suspended cells in rat bone marrowcells cultured for7days induced by GM-CSF, rmIL-4culture by inverted microscope, thenthere appears typical branch-like protrusions on the cell surface by plusing LPS andstimulating48hours, it completely accords with the form of a typical mature DC.(2)compared with control group, the expression of CD86、OX62respective is (61.51±0.89%)%、(85.20±1.41%), sinomenine group dendritic cell surface CD86, OX62positiveexpression rate of CD86expression (low concentration group (57.60±1.18%),(71.59±1.03%) OX62expression in CD86expression; concentration group40.59±1.08%theexpression of OX62,63.81±0.96%; high concentration expression of CD86、OX62group respective is (37.62±1.19%)(60.78±1.07%) expression difference was statisticallysignificant(p<0.05), indicating that the expression of sinomenine inhibits dendritic cells CD86, OX62.The CD86、OX62molecular positive expression rate of dendritic cell surfacein sinomenine group has statistical significance (P <0.05).(3) compared with LPS group,the positive expression rate of dendritic cell surface CD86(91.41±0.76%)、OX62(95.42±0.66%)molecular is significantly lowered, which intervened by differentconcentrations of sinomenine, the difference was statistically significant (P <0.05).Conclusion: the sinomenine can inhibit the dendritic cell surface CD86、OX62expression, thus inhibiting the mature dendritic cells.
