Objective To discover the relationship between the Helicobacterpylori (H. pylori) infection, Helicobacter pylori eradication using antibioticsand the diseases occurred in the lower esophagus, the changes of thebacterial flora in the lower esophagus in animal models of mice wereinvestigated.Methods Through established the BALB|C mouse models. A total of45BALB|C mouse were divided into3groups,blank control,infectiongroup and treatment group. The animal infection models and treatmentgroup were established through filling and feed the bacterial liquid ofHelicobacter pylori. After4weeks of the last filling and feed, the treatmentgroup were filled with Omeprazole, Ampicillin, and Clarithromycin.All the mice were put to death and the lower esophagus in mice weretaken. The DNA of bacteria was extracted in the lower esophagus and theV6areas in16S rDNA were amplified using the technology of polymerasechain reaction-denaturing gradient gel electrophoresis. The bacterial floras were analyzed on the DGGE atlas with Quantity-One1-D Analysis software.After the difference stripes in the three groups were amplified by the16SrDNA v6areas primer and their DNA were sequenced. The DNA sequenceswere compared and analyzed through BLAST.Results The H. pylori infection rate of infection group was100%.The H. pylori eradication rate of the treatment group was93%.DGGEfinger-prints showed that there was obvious differences (P<0.05) in thenumbers of bands among the three groups. The diversity index and richnessindex in the infection group were significantly different from negativecontrol group(0.01<P<0.05).The Evenness index was not significantdifference(P>0.05).The analysis of bacterium cluster class can be wellseparated in infection group dendrogram. The analysis of the principalcomponent showed that the different groups of bacteria were gathered in thedifferent positions. The special bacteria were found in the infection groupand the blank control group through the comparison of BLAST.Conclusion The results showed that the Lactobacillus and theBacteroides were the predominant bacterial flora colonized in the loweresophagus, and the structure of bacterial flora would change to thepredominant bacteria of Staphylococcus,Acinetobacter and Bacteridiumafter the infection of H. pylori.
