| Objective:To detect the different expressions of GM130,14-3-3δ and integrin α3in normal gastric tissue,moderately differentiated gastric cancer and poorlydifferentiated gastric cancer and thus to explore its relationship with gastriccancer.Methods:The expressions of ＧＭ130,14-3-3δand integrin α3were detected bySABC immunohistochemistry in clinical’s collection fresh gastriccarcinoma tissues of84cases of patients with gastric cancer and the84cases of normal gastric tissues (from5cm above the edge of the gastrictumor tissues).We analyzed the expressions of proteins combine withclinical pathological data as well.The expressions of mRNA and protein ofＧＭ130,14-3-3δand integrin α3were detected by RT-PCR and westernblot.Results: In the cases of gastric cancer, the positive expression rates of ＧＭ1301,14-3-3δ2and integrin α33(88.1%,90.5%,95.2%) were significantlyhigher than those in normal gastric tissues (52.4%,27.4%,42.9%),theexpression of the above three indicators in poorly differentiated gastriccancer group are higher than the moderately differentiated gastric cancergroup.The difference was statistically significant（P1<0.001、P2=0.007、P3<0.001）.Spearman correlation analysis showed that positive correlationwere observed in ＧＭ130,14-3-3δ and integrin α3in gastric cancer（P＜0.05）.The expression of ＧＭ130,14-3-3δand integrin α3in gastric cancerwas obviously related with pathological differentiation and clinical stage（P＜0.05）,but the correlation to age, gender,tumor location,tumor sizeinfiltration depth or whether the lymph node metastasis were not found（P＞0.05）.The level of mRNA and proteins were higher in gastric cancergroup than normal gastric tissue group, poorly differentiated gastric canceris higher than moderately differentiated gastric cancer,and the differencewere statistically significant between the two groups（P＜0.05）.Conclusion:The abnormal expression of GM130may play an important role in theoccurrence and development of gastric cancer. |
