Objective:1.To observe the influence of Tetraspanin Tspan9 on migration and invasion of gastric cancer cell SGC7901;2.To observe the influence of Tspan9 on expression of integrin beta 1;3.To observe whether integrin beta 1 is involved in this progress.Methods:1.First,we detect the expression of Tspan9 protein of our previous transfected SGC7901 cells(T-SGC7901 cells)through lentivirus transfection.2.Second,we detect the influence of Tspan9 on SGC7901 cells’ migration and invasion ability by wound healing assay and transwell assay respectively.3.Third,we establish ITG/T-SGC7901 cells through Lipofectamine 2000,and detect the expression of integrin beta 1 protein.4.Last,we detect the variation of migration and invasion of ITG/T-SGC7901 cells by wound healing assay and transwell assay respectively.Results : 1.T-SGC7901 cells express more Tspan9 protein than controls(P < 0.05);2.Wound healing assay show that T-SGC7901 cells’ migration rate is lower compared with SGC7901 cells(P<0.05),transwell assay show that T-SGC7901 cells’ invasion ability is weaker compared with SGC7901 cells(P < 0.05);3.After transient transfection,the ITG/T-SGC7901 cells express more integrin beta 1 protein;4.Wound healing assay shows that ITG/T-SGC7901 cells’ migration rate increase compared with the control cells(P <0.05),transwell assay shows that ITG1/T-SGC7901 cells’ invasion ability is enhanced compared with the control cells(P<0.05).Conclusion and significance : The tetraspanin Tspan9 can inhibit the migration and invasion of gasrtric cancer cell SGC7901,which maybe through the downregulation of integrin beta 1.Our experiment strengthen the understanding of the relationship between tetraspanins and malignancy,and provide experimental basis for further research.
