Investigation On Quality Standards Of Diyuhuaijiao Wan

ObjectivePerfecting the pharmacopoeia and improving quality standards ofDiyuhuaijiao Wan.To establish a series methods to identify traditional Chinesemedicine in Diyuhuaijiao Wan such as Rhubarb,Scutellarea,Angelica,Citrusaurantium.To develope methods to determine the content of rutin in Huaijiaoand Huaihua.To develop methods to determine the contents of paeoniflorin,hesperidin,baicalin in Diyuhuaijiao Wan in the same time.MethodsTo select the right extraction of active ingredients in Diyuhuaijiao Wan andto optimize the constitution and mixture ratio of eluent.Rhubarb,Scutellaria,Ang-elica and Citurs aurantium in Diyuhuaijiao Wan should be identified under thebest condition by TLC.Using RP-HPLC to determine rutin and determinepaeoniflorin,hesperidin,baicalin of Diyuhuaijiao Wan under the same conditionby choosing the constitution,proportion and temperature of mobile phase.Thendo the methodological study.ResultsThe methods were established for the identification of Rhubarb,Scutellaria,Angelica,Citrus aurantium in Diyuhuaijiao Wan with TLC,the characteristicsidentification were distinctive.There were no specific spot in the test solution andbank solution.Rutin,paeoniflorin,hesperidin and baicalin showed good linearity at the ranges of0.0044mg·mL-1~0.2200mg·mL-1,0.0062mg·mL-1~0.0620mg·mL-1,0.0080mg·mL-1~0.0800mg·mL-1,0.0105mg·mL-1~0.1050mg·mL-1.TheRSD were1.55,1.35,0.51,1.14.The content of rutin in Diyuhuaijiao Wan was56.51mg/pill,paeoniflorin was10.17mg/pill,baicalin was275.22mg/pill and bai-calin was81.81mg/pill.ConclusionsThe methods established for TLC identification were handy,rapid andspecific.The RP-HPLC method was established to determine rutin inDiyuhuaijiao Wan,and to determine paeoniflorin,hesperindin and baicalin inDiyuhuaijiao Wan under the same condition.The method was found to bebetter,more comprehensive to control medicine’s quality.It is also can providesufficient theoretical scientific basis for medicine’s producing,marketing,usingand testing.
