Objective: The Ubiquitin-like with PHD and ring finger domains1(UHRF1) has been reported to be essential for cell proliferation and play acritical role in the development and progression of many humancarcinomas. However, its relationship with laryngeal squamous cellcarcinoma(LSCC) remain unclear. Therefore, the expression and clinicalsignificances of UHRF1in laryngeal squamous cell carcinoma(LSCC)will be investigated in our study. Methods: In the current study,60patients with LSCC were studied.UHRF1expression at mRNA and protein levels were detected byquantitative reverse transcriptase-polymerase chain reaction (qRT-PCR),immunohistochemistry (IHC) and western blot respectively in bothtissues of LSCC and corresponding adjacent normal larynx tissues. Results: qRT-PCR showed that the expression of UHRF1mRNA inLSCC tissues was significantly higher than that in adjacent normal larynxtissues(p<0.001); By immunohistochemistry and western blot, overexpression of UHRF1protein was found in78.3%(47/60LSCC tissues, while there was negative expression in adjacent normal larynx tissues. Furthermore, increased expression of UHRF1had remarkably positiverelationships with smoking (P<0.001),advanced T stage (p=0.005) andclinical stage (P=0.044), poor histologic differentiation (P=0.048),whilethere was no correlations between UHRF1and sex, age and lymph nodemetastasis (P>0.05).Conclusion: Overexpression of UHRF1may play an important role inthe development and progression of LSCC.
