ObjectiveBenzo(a)pyrene (BaP) is persistent organic pollutants (pops),whichwidely exists in air, water. BaP is a serious threat to human health. Innumerous studies, BaP is harmful to pregnancy, but the mechanisms is stillunclear. Successful pregnancy depends on the receptivity of endometrium,embryonic vitality and the invasion of trophoblast cells. Trophoblast cellsinvasive into endometrium moderately is an important factor of successfulembryo implantation. Existing research shows that BaP can change cellmigration and invasion ability, promote the occurrence and metastasis oftumors.The biological behaviour of the embryonic invasion is similar withthe process of tumor formation, but whether BaP affect the normal functionof trophoblast cells, has not been reported. The aim of this study is toinvestigate the effect and possible mechanism of benzo(a)pyrene (BaP) onthe invasion and imgration of trophoblast cell.MethodsHTR-8/SVneo cells were cultured with different concentrations of BaP.MTT and Flow cytometry were used to detect the effect of BaP on cellvability and apoptosis. According to this result, optimum concentrationwere choosed for further study. The invasion and migration of HTR-8/SVneo cell were observed after BaP treatment.The expression ofgenes which is related to migration and invasion were detected. At the sametime the Western blot experiments was used to test protein expression insignal pathway related to invasion and migration.Results:1BaP inhibited the migration and invasion of extravillous trophoblastHTR-8/SVneo cells in a dose dependent.2HTR-8/SVneo cells were cultured with different concentrations ofBaP. the expression of MMP-2, MMP-9and E-cadherin were rised, and theexpression of E-cadherin is in dose dependent.3After extravillous trophoblast HTR-8/SVneo cells were culturedwith different concentrations of BaP. We found the expression of MMP-2and MMP-9, E-cadherin is regulated by the MAPK signal pathway.To sum up, the BaP can change the the migration and invasion ofextravillous trophoblast cell HTR-8/SVneo, and which is mediated by theMAPK signal pathway.
