ObjectiveTo investigate the Effect of Kushenin Combined with Bone Mesenchymal StemCells(MSCs)on the Repair of Colon mucosa in Ulcerative Colitis Rats.MethodsMSCs were isolated from the male rats;40Female rats were divided into fourgroups randomly. Group A was normal control, and groups B,C and D were UCmodels. The rats in group A and B were injected with1mL saline via tail vein, whilethose in group C and D with1mL1×106MSCs suspension. In addition, group Dwas treaded with Kurarinone for2weeks. After two weeks, colon tissue sampleswere analyzed for histopathology, for expression of Y chromosome-specific geneSRY in the colon tissues by PCR technology and Fluorescent in situ hybridization,for levels of bFGF in colon tissue by immunohistochemical and Western blotting,and for expression of bFGF, TNFα and IL-1β by RT-PCR.ResultsWe found that SRY was expressed in group C and D. The expression of. TNFα,IL-1β and bFGF were low in group A,while the expression of TNFα, IL-1βB andbFGF in group B were increased(P <0.05); Compared with group B, TNFα andIL-1β expressions in groupC were significantly decreased while the bFGF wasincreased (P <0.05); Compared with group C, TNFα and IL-1β expressions in groupD were reduced while bFGF was increased (P <0.05). ConclusionsThese data suggest that MSCs can home into colon tissue of rats; andsignificantly reduce the inflammation in colon tissue and repair the colon mucosadamage, which might be related to its effectively decrease expression of TNFα andIL-1β in colon tissue, and increase expression of bFGF. Combination of kusheninand MSCs have synergistic effect in treatment with UC in rat.
