Objective To analyse the correlation between the expression of VEGF with t-PA and PAIexpression in plasma, aqueous humor and vitreous of patient with proliferative diabeticretinopathy(PDR), to investigate the effect of VEGF, t-PA and PAI in pathogenesis of PDR, andto inspire new treatment idea and strategy.Methods The concentration of VEGF, t-PA and PAI in40cases plasma,20eyes aqueoushumor and20eyes vitreous body in PDR group, the concentration of VEGF, t-PA and PAI in40cases plasma and19eyes aqueous humor in control group were detected by enzyme linkedimmunosorbent assay(ELISA). Observed the differences between the two groups and thecorrelation between VEGF and t-PA, VEGF and PAI by rank sum test and Spearman correlationanalysis.Results1. The comparison of VEGF, t-PA and PAI in plasma of PDR group and control group(1) The VEGF concentration in plasma of PDR group (53.37pg/ml) is higher than controlgroup (15.08pg/ml), and there was statistically difference between them (P<0.05).(2) The t-PA concentration in plasma of PDR group is24.32ng/ml, control group is12.53ng/ml,and there was no statistically difference between them (P>0.05).(3) The PAI concentration in plasma of PDR group (8.00ng/ml) is higher than control group (0.19ng/ml), and there was statistically difference between them (P<0.05).2. The comparison of VEGF, t-PA and PAI in aqueous humor of PDR group and control group(1) The VEGF concentration in aqueous humor of PDR group (335.00pg/ml) is higher thancontrol group (36.58pg/ml), and there was statistically difference between them (P<0.05).(2) The t-PA concentration in aqueous humor of PDR group (5.70ng/ml) is higher than controlgroup (3.68ng/ml), and there was statistically difference between them (P<0.05).(3) The PAI concentration in aqueous humor of PDR group (0.63ng/ml) is higher than controlgroup (0.11ng/ml), and there was statistically difference between them (P<0.05).3. The comparison of VEGF, t-PA and PAI in plasma, aqueous humor and vitreous of PDRgroup(1) The comparison of VEGF: the comparison of vitreous (691.69pg/ml) is higher thanaqueous humor (335.00pg/ml), and there was statistically difference between them (P<0.05),aqueous humor is higher than plasma (53.37pg/ml), and there was statistically differencebetween them (P<0.05), the vitreous is higher than plasma, and there was statistically differencebetween them (P<0.05).(2) The comparison of t-PA: the comparison of plasma(24.32ng/ml) is higher than vitreous(13.05ng/ml), the vitreous is higher than aqueous humor (5.70ng/ml), the plasma is higher thanaqueous humor, and there were statistically difference between them (P<0.05).(3) The comparison of PAI: the comparison of plasma and vitreous was no statisticallydifference between them (P>0.05), the comparison of plasma (8.00ng/ml) and vitreous(4.01ng/ml) are higher than aqueous humor (0.63ng/ml), and there was statistically differencebetween them (P<0.05).4. The correlation between the expression of VEGF with t-PA and PAI expression in plasma,aqueous humor and vitreous of PDR(1) In plasma: the expression of VEGF and t-PA is significant correlated (r=0.659, P<0.05);the expression of VEGF and PAI is significant correlated (r=0.705, P<0.05). (2) In aqueous humor: the expression of VEGF and t-PA is significant correlated (r=0.572,P<0.05); the expression of VEGF and PAI is significant correlated (r=0.648, P<0.05).(3) In vitreous: the expression of VEGF and t-PA is significant correlated (r=0.798, P<0.05);the expression of VEGF and PAI is significant correlated (r=0.694, P<0.05).Conclusion1. VEGF, t-PA and PAI were may involved in the process of PDR retinal neovascularization.2. VEGF play to stimulate the growth of neovascularization mainly in the eye local, to detectthe concentration of VEGF in plasma does not as the monitoring indicators of growth of ocularneovascularization.3. PDR patients with blood fibrinolysis disorders, t-PA and PAI as a key part of the bloodfibrinolytic system is also involved.4. VEGF and t-PA, PAI may have a mutual promotion role in the development of the PDR.
