Background and Significance Pituitary adenomas are common benign intracranial neoplasms representing about 10-25% of all intracranial neoplasm. Significant morbidity can occur along with pituitary adenomas due to hormonal dysfunction and mass effects. The pathogenesis of pituitary adenoma is unclear. Pituitary adenoma is divided into invasive pituitary adenoma and non-invasive pituitary adenoma by biological behavior. In clinical, invasiveness is the major factor that determines therapeutic efficacy and prognosis in pituitary tumors, including the fate of postoperative residual tumor, rate of remission, rate of recurrence and patient survival. MicroRNAs (miRNAs) are small non-coding RNA molecules that represent a major class of molecular regulators. miRNAs have been implicated in the pathogenesis of several human tumors, including pituitary adenomas. Altered expression of miRNAs has been describedin pituitary adenomas, and specific miRNA signatures are related to clinical and therapeutic characteristics of the tumors. Recently, it has been found that miRNAs also stably express in serum, plasma and other body fluids. Moreover, the expression levels of some circulating miRNAs specifically change in patients with certain diseases.Objective First finding the differentially expression of some miRNAs in tissues and serum, from the patients diagnosed as invasive pituitary adenoma were compared with non-invasive pituitary adenoma, and then finding out the significant miRNAs which are both in tissues and blood. To determine whether miRNAs can be used for an indicator to judge the pituitary adenoma is invasive or not.Methods Collecting the patients diagnosed as pituitary adenomas, according to Hardy-Wilson grade, the patients were divided into two group which are the typical invasive pituitary adenoma and the typical non-invasive pituitary adenoma, each group has 5 cases, first using the miRNAs genechip to detection the tissues and serum from the invasive pituitary adenoma patients and non-invasiv pituitary adenoma patients. Then using the method of fold change to select the differentially expression of miRNAs whose the fold change are larger than 2 and equal to 2 or lesser than 0.5 and equal to 0.5 as the significant miRNA from two groups. Through the computer simulateon to find out the miRNAs existing both in tissues and serum, and predict the target genes of the miRNAs. The last is proving the results that is consistent by RT-PCR method.Results The differentially expression of miRNAs are 388 in tissues,62 in serum,10 miRNAs are existing both in tissues and serum. From the 10 miRNAs, we found that a few of genes predicted the target genes of has-miR-142-3p, supplied by miRGen, have been reported to participate in tumorigenesis and cancer progression. Then using RT-PCR proving the results statistically and significant.
