Objective: Tumor associated macrophage(TAMs) and Her-2 play an extremely important role in the process of tumor progression. The aim of the research was to explore the protein expression of TAMs and Her-2 in the breast carcinoma tissue and adjacent tissue by SP immunohistochemistry, to analyze the significance of TAMs and Her-2 protein expreessionn in the breast cancer tissue.Methods:1 104 specimens from patients with breast cancer patients were operated in Hebei Province People’s Hospital, All the patients did not accept radio-therapy,chemical-therapy and endocrino-therapy.To detect the expression of TAMs and Her-2 in carcinoma tissues and adjacent tissues by SP immunohistochemistry.To study the relation between TAMs and Her-2 and the clinic pathology’s parameter of breast carcinomas.2 Immunohistochemistry examin:TAMs(CD68): The staining intensity and the number of positive cells proportion were scored as follows: 0= no staining or 0-5%; 1= light yellow granules(weak),or 1= 5-25%; 2= dark yellow(medium),or 26-50%; 3=brown granules(strong),or 51-75%. The labeling score defined by plus the percentage of positive cells and the staining intensity was the final score for the section. When the total score 0-1 was(-), 2-3 was(+),4-6 was(++),>6 was(+++),and then(-)-(+) was defined as negative;(++)-(+++) was defined as positive. Her-2:The staining intensity was scored as follows:(-)=no staining,or <10%tumor cells appear very weak membrane staining;(+)=light yellow granules or >10%tumor cells appear very weak cell membrane;(++)=weak-medium intensity cell membrane or >10%tumor cells appear;(+++)=strong cell membrane or >10%tumor cells appear. When the total score was(-)-(+),the case was defined as positive. When the total score was(++)-(+++), the case was defined as negative.3 Statistical handling used SPSS 13.0 software. We used the chi-square test for the comparision of the expression in every tissue and the relation between CD68 and Her-2. It used the spearman rank test for the correlation between the expression of CD68 and Her-2. P < 0.05 were considered distinct difference.Results:1 The significance of TAMs in Breast Cancer:The expression of CD68 in the different tissues:The carcinoma tissues was 73.1%(76/104), The adjacent tissues was 14.4%(15/104). In this two tissues,the expression of CD68 has distinct difference(P<0.05).The positive expression of CD68 has distinct difference with differentiation degrees, tumor size, TNM stage, lymph node metastasis, blood vessel invasion(P<0.05). While there was no distinct difference with age(P>0.05). The positive expression of CD68 in patients with highly、moderatly and poorly differentiation degrees was 65.7%(46/70),88.2%(30/34), with tumor size ≤ 2cm, > 2cm was 52.6%(20/38),84.8%(56/66); withⅠ-Ⅱ stage, Ⅲ-IVstage was 63.6%(35/55), 83.7%(41/49); with lymph node metastasis,without lymph node metastasis was 86.4%(38/44), 63.3%(38/60); with blood vessel invasion, without blood vessel invasion was 84.8%(39/46),63.8%(37/58).2 The significance of Her-2 protein in Breast cancer:The expression of Her-2 in the different tissues: The carcinoma tissues was 57.7%(60/104),The adjacent tissues was 9.6%(10/104). In this two tissues,the expression of Her-2 has distinct difference(P<0.05).The positive expression of Her-2 protein has distinct difference with differentiation degrees, tumor size,TNM stage, lymph node metastasis, blood vessel invasion(P<0.05). While there was no distinct difference with age(P>0.05). The positive expression of Her-2 protein in patients with highly、moderatly and poorly differentiation degrees was 48.6(34/70),76.5%(26/34),with tumor size ≤2cm、>2cm was 31.6%(12/38),72.7%(48/66); withⅠ-Ⅱ stage, Ⅲ-IVstage was 41.8%(23/55),75.5%(37/49); with lymph node metastasis, without lymph node metastasis was 72.7%(32/44), 46.7%(28/60); with blood vessel invasion, without blood vessel invasion was 78.3%(36/46),41.4%(24/58).3 The relationship between expression of TAMs and Her-2 in breast carcinoma tissues:In 104 cases of patients, the cases of CD68 and Her-2 double positive was 38, CD68(+) while Her-2(-) cases was 22, CD68(-) while Her-2(+) cases was 38, the cases of CD68 and Her-2 double negative was 6. There was distinct difference between groups(c2=6.843,P<0.05). The Spearman rank test, between CD68 and Her-2 in the breast carcinoma tissues were positively correlated(r=0.257).Conclusions:1 The positive expression of TAMs(CD68) was significantly higher than adjacent tissues in breast carcinoma tissues; The positive expression of Her-2 was significantly higher than adjacent tissues in breast carcinoma tissues. The expression of TAMs(CD68) and Her-2 were positively correlated in the breast cancer tissues.2 The expression of TAMs(CD68) and Her-2 in carcinoma tissues were no distinct difference with age. While there was distinct difference with differentiation degrees, tumor size, TNM stage, lymph node metastasis, blood vessel invasion in breast carcinoma patients.
