Poria cocos is the arid sclerote of Poria cocos(Sch.Wolf)which is a kind of mycetes belonging to Polyporaceae.It is admitted in Chinese Pharmacopeia(2005).Shengnong's herbal regards Poria cocos as high grade medicine,which can excrete dampness,use diuretic of hydragogue to alleviate water retention,invigorate the spleen and calm down the cardia. Lots of documents report the major chemical compositions are polysaccharides and triterpens.Modern pharmacology research on poria cocos show that it can significantly regulate immune function,repress tumor,has anti-inflammatory function and so on.The material basis of Poria cocos' pharmaco-function is researched on in this paper,while the bioactivity of polysaccharides(degraded polysaccharides and carboxymethyl-polysaccharides),the alcohol extracted,the acetic ether extracted,n-butanol extracted and the remaining water is sieved.Tracing the pharmacological results,the active parts and the major chemical compositions are ascertained.By using phenylic alcohol sulfuric acid,the content of polysaccharides is determined.Based on inspection of effect of granulation,mediun,method and time on the extraction,orthogonal design is used to optimize extraction with lye.Degraded polysaccharides is obtained with Smith degrading method.Carboxymethyl-polysaccharides is obtained with twice lying.Considerating the react state,medium,time, temperature,material ratios and so on,the purity and the characters are under control.The reactive condition and cost is selectively optimized. Soluble carboxymethyl-polysaccharides is gained at last,and the D.S is 0.8.6 compounds are separated from poria cocos,and 5 of them are identified.They are ergosterin(Ⅰ),poria acid(Ⅱ),oleanolic acid(Ⅲ), dehydroeburic acid(Ⅳ)and 3β-hydroxy-lanostane -7,9(11),24-triene -21-acid(Ⅴ).Comparing research on fingerprint for the acetic ether extraction from different habits with HPLC,10 common summits are identified,and all the summits are separated in 90 mins.All the fingerprint paraments accord with correlated requirements and the method has good reproducibility and constancy.This research provides scientific certain for assessment of poria cocos and establishs foundation on the quanlity control of poria cocos.
