[Objective] To quantify relatively the gene expression of fatty acid synthase in squamous cell carcinoma , adjacent tissue ,and some normal oral tissues by real-time quantitative PCR., which is an important contribution to the diagnosis of the squamous cell carcinoma of oral cavity and the choice of clinical treatment by fatty acid synthesic pathway.[ Methods ] Twenty patients who pathologically proved squamous cell carcinoma of oral cavity were studied. The tissues were collected fresh from surgical specimens. The collected tissues were minced. Then the total RNA was extracted .The RNA was reversely transcripted into cDNA with randomprime. And then Taqman probe and primers were added to perform real-time quantitative PCR in GeneAmp PCR System FTC2000 sequence detection system to quantify the gene expression of FAS according to an internal control G A P D H . FAS and G A P D H were as target gene and internal reference respectively. Standard curves were established using a series dilution of quantificated palsmids to measure the amplification efficiency of FAS and G A D P H. Ct value of FAS and GADPH were measured in 20 samples stimuteneously. The difference of F A S gene expression was compared between squamouscell carcinoma tissues, adjacent tissues, and some normal oral tissues by the AACt and 2 AA" methods .[Results] The FAS mRNA levels in adjacent tissues were 6.450 + 1.063, significantly higher than those in normal oral tissues(7.625 ± 1.223) and lower than squamous cell carcinoma tissues(5.000 ± 1.0760).(t = 7. 012 p= 0. 041 and t= 6. 766 p= 0. 035). The differentiation degree of squamous cell carcinoma is not relative to the FAS gene expression.(p>0. 05) .[Conclusion] The expression of FAS mRNA is hightest in squamous cell carcinoma tissues, moderate in adjacent tissues, and lowest in some normal oral tissues. Real-time quantitative PCR provides a method for monitoring the expression of fatty acid synthetic activity in squamous cell carcinoma, adjacent and normal tissues .The technique has fine sensitivity and reproducibility and the data analysis was simple and reliable and contributes to the choice of clinical treatment, which has a bright future in application .
