For wound involving injury to dermis, formation of pathological scar is an inevitable result. How to effectively promote wound scarless healing has been a world problem. Studies have showed that the healing speed of wounds in moist environment is twice times in the dry environment. Therefore, the FDA issued medical topical wound dressing guidelines of industry emphasize that keep the wound moist environment is an important method for standard treatment. Under the guidance of the theory of "moist wound healing", functional medical dressings with the advances in biological medicine and materials science have undergone revolutionary changes. However, the current commercially available wound dressings are of single function, using chemical antimicrobial drug, which readily causes to further injury to the wounded tissue cells, large amount of tissue fluid leakage, inflammation, and eventually formation of pathological scar.In this study, we used chitosan and carboxymethyl-chitosan as carrier material, recombination with Notoginseng extract to make the new wound gel. Use high-performance liquid chromatography to establish method for determination of ginsenosides Rgi and ginsenoside Rb1 in dressing and the methods are verified by experiment. Results showed that in the concentration range of 62.5-1000 μg/mL, regression equation of ginsenosides Rg1 and Rb1 are Y=2250.3X-3265.8 (R2=0.9998, n=5) and Y=2521.5X+112187 (R2=0.9999, n=5), which have good linearity; precision is good,1.60%(n=6) and 1.61%(n=6); both recoveries in the range of 90.0-110.0%, the mean value is 99.7% and 101.2%. This method is fast and accurate, and can be used in gel production quality control.Notoginseng extract was prepared by alcohol reflux, and Chitosan lactate, o-carboxymethyl Chitosan and Notoginseng extract were chosen as the prescriptions of chitosan hydrogel dressing. Through water testing, contact stability test and determination of dressings shrinkage experiments, determine the proportion of prescriptions is 4:1:5. Hydrogel-release release data in accordance with first level prescription process, active ingredients in cumulative release rate of 97.4% in 2 h, and 3 h can be fully released, indicating that gel rapid can release of active ingredients acting on the wound. After stability test, the character and content of the product didn’t change significantly, indicates that products prescription, preparation technology and choice of packaging material are reasonable, product stability is good.Hemolysis test was done on products, and the total cortical excision wound model to promote rapid scarless wound healing effects of rabbits’ skin wound were evaluated. Observe wound healing of burn patients and patients with incision after applying the gel dressing. Results showed that the gel does not cause hemolysis; can form a gel layer over the wound and absorb wound exudate; quickly stop bleeding, reduces exudation, moisture, ventilation, anti-inflammatory, antipruritic, adhesions and other advantages; can accelerate wound healing, inhibit scar formation, reaches the ideal functional dressings required for external use.
